摘    要:目的:探讨P物质(SP)对嗜酸性粒细胞(Eos)粘附功能的调节作用及机制。方法:用收集^51Cr标记的白细胞分析法观察人Eos对培养的人脐带静脉血管内皮细胞(HUVEC)的粘附作用。在实验中以不同浓度SP和SP+血小板激活因子(PAF)分别预处理Eos,观察其对经IL-1β活化的HUVEC粘附性变化。以GR71251(NK-1受体阻断剂)和抗CD11b作为干预因素,观察它们对SP+PAF物影响。

Abstract:AIM: To investigate the effects of substance P(SP) on the human eosinophil adhesion to endothelial cells and its mechanism. METHODS: Eosinophil adherence to human umbilical vein endothelial cells (HUVEC) was measured by using a 51Crradiolabeled leukocyte collection assay. In the experiment, we observed the influence of SP, combination of SP and platelet activating factor (PAF) on the human eosinophil adherence to HUVEC, respectively. Additionally,we observed whether GR71251, NK1 receptor specific antagonist, and antiCD11b would influence the effect of SP. Adhesion of SPtreated HUVEC to human eosinophil activated by SP and PAF was also observed in this study. RESULTS: SP did not increase the adhesion of human eosinophil to the HUVEC activated with IL1B alone but could significantly enhance the upregulative effect of PAF on adhesion of human eosinophil to HUVEC activated by IL1B.GR71251 and antiCD11b could inhibite the effect of SP respectively (inhibition rate: 29.2% and 59.2%, respectively). The HUVEC activated by SP showed increased adhesion to the eosinophie activated by SP and PAF. The effect of SP on adhesion of HUVEC to eosinophil was dosedependent,and inhibited by GR71251. CONCLUSION: SP can enhance the upregulative effect of PAF on the eosinophil adherence to HUVEC. NK1 receptor and CD11b may be involved in the effect of SP. SP activates the HUVEC, and increases the adhesion of HUVEC to eosinophil activated by SP and PAF.
