摘    要:目的:探讨通过RNA干扰技术下调SOCS3表达防治高脂餐大鼠代谢紊乱的作用.方法:取SD6wk龄大鼠45只,随机分为3组:普通饮食对照组(n=15),下丘脑注射空白质粒,普通饲料喂养; 高脂饮食对照组(n=15),下丘脑注射空白质粒,高脂饲料喂养; 实验组(n=15),下丘脑注射shR-NA-SOCS3序列慢病毒重组体,高脂饲料喂养.每周测体质量1次.饲养8wk末,测空腹血糖和体质量,处死后立即取心脏血进行血清胰岛素、三酰甘油、总胆固醇、血清瘦素测定.结果:实验组体质量、三酰甘油、总胆固醇低于高脂饮食对照组(P〈0.01),高于普通饮食对照组(P〈0.05).实验组血清瘦素、血清胰岛素浓度低于高脂饮食对照组组(P〈0.01),与普通饮食对照组比较无统计学差异.结论:下调SOCS3基因表达可以降低体质量、三酰甘油、血清胰岛素、总胆固醇及血清瘦素水平,对肥胖症、糖尿病及慢性并发症可能有预防和治疗作用.
