糖尿病大鼠海马胆碱乙酰转移酶表达与认知功能的相关性

摘    要:目的:观察糖尿病大鼠海马胆碱乙酰转移酶(ChAT)mRNA和蛋白表达及其与认知功能的关系,探讨糖尿病脑病的发病机制.方法:38只SD雄性大鼠随机分为正常对照组(C组)和糖尿病组(D组),腹腔注射链脲佐菌素建立糖尿病大鼠模型.建模成功后11wk用Morris水迷宫测试大鼠学习和记忆能力;RT-PCR,原位杂交法检测ChATmRNA表达;免疫组化,WesternBlot法检测ChAT蛋白表达.结果:D组大鼠学习和记忆能力明显减退,其逃避潜伏期时间延长;C组40.90±10.90与D组77.56±27.86相比较,差异具有统计学意义(P〈0.05).穿越目标区域次数C组(3.93±0.44)次与D组(1.37±0.85)次相比较明显减少(P〈0.05);中心区域停留时间百分率C组(5.41±0.97)%与D组(2.20±1.28)%相比较明显下降(P〈0.05).海马ChATmRNA和ChAT蛋白表达C组0.48±0.03,0.55±0.02与D组0.37±0.01,0.33±0.01相比较均明显降低(P〈0.05).结论:糖尿病大鼠海马ChATmRNA和蛋白低水平表达可能是糖尿病脑病的发病机制之一.
