摘    要:目的 研究p16基因和顺铂联合应用对细胞系QBC939裸鼠皮下移植瘤模型的治疗作用。方法 将重组体腺病毒p16(Ad-p16)和顺铂联合作用于胆管癌细胞系QBC939裸鼠皮下移植瘤,观察和分析其对肿瘤的生长抑制作用。结果 用Ad-LacZ进行重组体腺病毒转导效率的检测,发现当感染强度在100MOI(Multiplicity of infetion)以上时,Ad-p16可使90.0%以上的培养的人胆

Abstract:AIM To evaluate the inhibitory effect of recombinant adenoviruses p16 (Ad-p16) with cisplatin (CDDP) on the nude mice model bearing subcutaneous tumor of cholangiocarcinoma cell line. METHODS The effects of Ad-p16 with CDDP on the growth of QBC939 cell in vivo were observed. RESULTS The QBC939 cell line transducted with recombinant adenoviruses LacZ (Ad-LacZ) at MOI of 100 or greater exhibited more than 90.0% transduction rate. Experimental therapy on the nude mice model bearing subcutaneous tumor of QBC939 cells showed that intratumor instillation of Ad-p16 and in celiac instillation of cisplatin (CDDP) inhibited the growth of the tumors. The average weight of the Ad-p16-treated, CDDP-treated or Ad-p16 with CDDP-treated tumors were inhibited 30.0%, 41.0% or 62.6% respectively, compared with that of the tumors injected with Ad-LacZ. CONCLUSION p16 gene can increase the chemosensitivity of cisplatin in human biliary duct cancer cell.
