人胃癌裸鼠胃原位移植转移模型的建立

目的 :建立人胃癌裸鼠原位移植转移模型 .方法 :用人胃癌细胞悬殊液接种于裸鼠皮下 ,形成稳定传代的皮下移植瘤 ,将皮下移植瘤组织再接种于裸鼠胃浆膜下 ,称为间接胃原位移植模型 ,并与直接用细胞悬液接种于裸鼠胃浆膜下、皮下、腹腔、尾静脉的移植瘤模型比较 ,观察移植肿瘤的生长状况、移植成功率和自发转移的发生率及组织学变化及细胞动力学改变等生物学特性 .结果 :肿瘤组织块保留了人胃腺癌的组织学特片 ,尾静脉注射与直接原位接种成瘤率均为6 0 % ,间接原位接种模型移植成瘤率为 80 % ,皮下与腹腔接种成瘤率均为 10 0 % .间接胃原位移植模型肝脏转移率为6 0 % ,直接胃原位移植模型肝脏转移率为 33.3% (P <0 .0 5 )尾静脉移植瘤模型仅见肝脏转移 ,实验过程中裸鼠衰竭处死后的肺脏经连续组织切片未见瘤细胞侵袭 .移植瘤细胞具有分泌CEA的功能 ,细胞动力学检测显示以五倍体细胞为主 .结论 :人胃癌裸鼠皮下 -胃原位移植瘤模型的生物学行学与人胃癌自然生长和转移过程相似 ,为研究人胃癌转移机制提供了理想的模型 .

Establishment of transplantation of in situ human gastric tumor metastasis model in nude mice

AIM: To establish an orthotopic transplantation gastric tumor model in nude mice. METHODS: Tumor cell line of human gastric adenocarcinoma was inoculated subcutaeously into nude mice to develop implant tumor, then intact tumor tissue was collected and implanted orthotopically into the nude mice stomach, the model was named indirect gastric orthotopic transplantation. Comparison of tumor growth, the rate of tumor survival and spontaneous metastasis with direct orthotopic transplantation model, subcutaneous model, peritoneal cavity model and tail vein model using cell suspensions was carried out. The histological characteristics and cell DNA content of transplantated tumors were also investigated. Flow cytometer was employed to detect tumor cell ploidy. RESULTS: The translantation tumors resemble the original human gastric adenocarcinomoa histologically. Tumor take rate of both tail vein model and direct orthotopic model were 60.0%. tumor take rates of both subcutaneous and peritoneal cavity model were 100%; the liver metastases rate of indirect and direct orthotopic transplantation were 60.0% and 33.3% in the nude mice respectively ( P <0.05).Only liver metastasis was observed in the model of tail vein injection. The histology of lungs was examined via serial sections after nude mice exhausted, but no metastasis was observesd during the experiment. With flow cytometer the cells were mainly five ploidy. Tumor cells can excret CEA. CONCLUSION: In the growth and metastasis orthotopic transplantation model is similar to human gastric cancer.
