摘    要:为探讨c-erbB-2基因对细胞增殖的影响。本实验通过构建c-erbB-2基因的表达载体一转染NIH3T3细胞,观察了转染细胞的生长特性。发现其对数生长期提前,群体倍增时间缩短,并且其克隆形成率,软琼脂集落形成能力,及裸鼠致瘤能力均明显高于对照组。

Abstract:Objective: In order to investigate the influences of c-erbB-2 gene on cell biological properties. Methods: The c-erbB-2 gene expressing vector was constructed and transfected into the NIH3T3 cell line, and then growth properties of the transfected cell line erbBTr were observed. Results: The cell lines achieved the following properties: 1. earlier logarithmic growth phase and shorter time-span of multiplication (39 h, P<0. 05). 2. higher clone forming rate in culture media (26% , P<0. 05) and in soft arga (17 clones/cell, P <0. 05). 3. stronger ability of induced tumors in nude mice (3/5). Conclusion; It can be deduced that the c-erbB-2 gene has some neoplastic transforming ability.
