大鼠肝细胞原代长期培养的形态学观察

作利用Ｉ型胶原酶灌注雄性ＳＤ大白鼠肝脏制备游离肝细胞悬液，以ＤＭＥＭ培养基进行培养观察。实验中观察到经多次低速离心后，细胞悬液中各种杂质基本消除。接种培养１２ｈ可见肝细胞贴附、伸展，呈圆形、椭圆形及不规则多边形，大部分肝细胞相互聚集，排列成簇或条索状，其中多见双核肝细胞。培养７ｄ后可见新生肝细胞呈椭圆形及多边形。培养５１ｄ时增殖的肝细胞数量明显增加，仍为多边形，相嵌紧密排列，此时可见其特有的肝细

Morphologica1 observation of long-term primarily cultured rat hepa- tocytes

In this study,We prepared the hepatocytes by perfusing the rat liver with collagenase and culturedthem with DMEM media.We observed the cell morphology by light and electron microscopy and found allsorts of impurities in cell suspension eliminated after low speed centrifugation.After culture for 12 h,hepa-tocytes were found to attach and extend.They were round,elliptic and polygonal in shape.Most hepatocytesaggregated and arranged in group or cord.Some of them were binuclear.After culture for 51 d,the prolifer-ative hepatocytes increased in number,they were still polygonal.They were arranged compactly.Then wecould see the particular ultrastructure of the hepatocytes:bile canaliculi,tight junction,glycogen rosettesand wheel- shaped mitochondria.Their ultrastructure was the same as that of the hepatocytes cultured for 2d.
