当归多糖硫酸酯的合成及其对脾细胞增殖的作用

摘    要:目的:合成不同取代度的当归多糖硫酸酯;探讨氯磺酸对当归多糖硫酸酯化过程的影响和当归多糖及其硫酸酯对小鼠脾细胞增殖反应的作用。方法采用氯磺酸-吡啶法进行硫酸化修饰;BaSO4比浊法测饰物的硫含量;MTT比色法研究当归多糖及其硫酸酯对小鼠脾细胞增殖反应的影响。结果得到8种不同取代度的当归多糖硫酸酯,硫含量为93.7-171.4mg.g^-1,产率为0.737-0.884g.g^-1,当归多糖及其硫酸酯在剂量为500mg.L^-时,给药组光密度明显高于对照组(P<0.01),作用与剂量有良好的正相关性。结论提高氯磺的比例,可以提高产物的硫含量,但收率降低,当归多糖及其硫酸酯对活化的T细胞增殖有直接促进作用。

Abstract:AIM To synthesize Angelica Sinensis polys accharide sulfates with diff erent sulfur content; to explore the effect of chlorosulfonic acid on sulfation in an excess of pyridine and the action of Angelica Sinensis polysaccharide sulf ates (APSs) on mouse splenocyte proliferation. METHODS  Angelica Sinensis polysa ccharide (AP-Ⅰ) was sulfated by chlorosulfonic acid-pyridine method; the este r sulfate was estimated turbidimetrically as barium sulfate; the effects of APSs on mouse splenocyte proliferation were studied by MTT assays in vit ro. RESULTS Ei ght products with different sulfur content were obtained. The yield was 0.737 ~0.884 g*g-1,and the sulfur content was 93.7~171.4 mg*g-1 . Both AP-Ⅰ and APSs enhanced m ouse splenocyte proliferation at dose 500 mg*L-1 and 1500 mg*L-1 ( P<0.01). CONCLUSION By increasing the ratio of chlorosu lfonic acid, sulfur content can be en hanced, but the yield will be reduced. AP-Ⅰand APSs possess direct promoting effects on active T cell proliferation.
