柯萨奇B3病毒抗原决定簇的多态性和致病性

摘    要:目的分析由各脏器分离出的柯萨奇B3病毒(CVB3)抗原决定簇的异同,并探讨其与致病性的关系. 方法利用自制的26种柯萨奇B3病毒mAb(CVB3- McAb)与心脏、肝脏、肾脏及肺脏分离出的CVB3株进行免疫荧光分析,观察各分离株抗原决定簇的变化,并将各分离株再次注入动物体内,观察心、肝、肾及肺的病理改变. 结果 CVB3在不同器官的细胞中繁殖后,其抗原决定簇发生改变, CVB3-McAb 2A4, 2B2, 2D3, 2D4与肝、肺、心、肾分离株均发生反应;26种CVB3-McAb中有4种不与心脏分离株反应;有15种不与肝脏分离株反应;有21种不与肺脏分离株反应;有11种不与肾脏分离株反应. 且各分离株对器官的致病程度亦有差异. 结论 CVB3各分离株抗原决定簇有差异;病毒抗原决定簇的不同是影响其致病性的重要因素之一.

Abstract:AIM To analyze antigenic determinant of Coxsackie virus B 3 (CVB 3) separated from different organs and to explore its relationship with pathogenicity. METHODS Immunofluorence analysis of CVB 3 strains separated from heart, liver, kidney and lung with 26 strains of our self making Coxsackie B 3 monoclonal antibody (CVB 3 McAb) was performed, and the alteration of antigenic determinants of the isolates was observed. We injected the isolates into animalbody again and observed the pathologic alteration of heart, liver, kidney and lung. RESULTS After the virus reproduced in different organs, Coxsackie B3 viral antigenic determinant changed, and CVB 3 McAb2A 4, 2B 2, 2D 3, 2D 4 all reacted with the isolates of heart, liver, kidney and lung. 4 of 26 strains did not react with isolates of heart, and 15 not with isolates of liver, 21 not with isolates of lung and 11 not with isolates of kidney. The pathogenic toxicity of the isolates to organs was also different. CONCLUSION The CVB 3 isolates are different in antigenic determinant, and the difference of viral antigenic determinant is one of the important factors that affect the pathogenicity.
