摘    要:目的 探讨小干扰RNA沉默HO-1基因表达后食管鳞癌Eca109细胞对氟尿嘧啶化疗敏感度的影响.方法 利用HO-1小干扰RNA(siRNA)及氯化高铁血红素分别沉默和诱导食管鳞癌Eca109细胞HO-1基因的表达,RT-PCR检测HO-1 mRNA水平,Western blot法检测HO-1蛋白表达水平,分别应用MTS法、流式细胞仪检测干预后Eca109细胞对氟尿嘧啶敏感度的变化.结果 RT-PCR、Western blot法检测结果显示,HO-1 siRNA能够有效抑制食管鳞癌Eca109细胞HO-1的表达.HO-1基因干扰后,氟尿嘧啶对Eca109细胞增殖抑制作用增强、凋亡细胞比例增加.结论 干扰HO-1的表达可以显著增强食管鳞癌Eca109细胞对氟尿嘧啶化疗的敏感度.

Interference of Heme Oxygenase-1 Gene Expression Enhances the Sensitivity of Esophageal Squamous Cell Carcinoma Eca109 to Fluorouracil

Abstract:Objective To explore the effect of sensitivity of esophageal carcinoma cell line Eca109 to fluorouracil by RNA interference suppressing the expression of HO-1.Methods The small interfering RNA (siRNA) targeting HO-1 gene was designed and synthesized to suppress the gene expression of HO-1,and hemin was used to promote the expression of HO-1.The mRNA and protein expression levels of HO-1 were evaluated by RT-PCR and Western blot.The effect of sensitivity of esophageal carcinoma cell line Eca109 to Fluorouracil was detected by MTS and flow cytometry.Results RT-PCR and Western blot test results showed that Hemin(10μmol/L) could effectively induce the expression of HO-1,while HO-1 siRNA could successfully inhibit the expression of HO-1 in esophageal squamous cell line Eca109.The proliferative capacity was markedly decreased and apoptosis index significantly increased after transfection with HO-1 siRNA.Conclusion The sensitivity of esophageal squamous cell line Eca109 to fluorouracil could be significantly improved by RNA interference suppressing the expression of HO-1.
