氯哌酰心安-5心血管药理作用研究

摘    要:氯哌酰心安-5是新的α、β受体阻断剂,大鼠iv7.5、11.4mg/kg能降低血压、减慢心率并抑制内脏交感神经放电,抑制兔主动脉环由去甲肾上腺素引起的收缩,减慢豚鼠离体心房的自发频率并使异丙肾上腺素加快心房自发频率的量效曲线右移,降低垂体后叶素引起的心电图ST段的升高,对麻醉大鼠血液动力学呈现抑制性影响,小鼠iv LD_(50)为49.5mg/kg。

Abstract:Chlopinolol, a new alpha-and beta-blocking agent, was synthesized in China. Chlopinolol 7.5,11.4 mg/kg iv markedly reduced blood pressure, heart rate and the splanchnic sympathetic nervedischarges (SND) in rats. It has been shown to have inhibiting effect on the contractile responeses ofisolated aorta rings in rabbit or contrations induced by noradrenaline. It was found that Chlopinololmarkedly inhibited the rate of spontaneous beating of guinea-pig atria or beat induced by isoprenaline.It reduced the ST segment of ECG induced by iv pituitrin. The effects of Chlopinolol iv administra-tion on haemodynamic were studied in anasthetized rats. It reduced LVP, LVdp/dt_(max) and heart rate.LD_(50) was 49.5 mg/kg in mice.
