复方丹参联合阿司匹林对大鼠糖尿病肾病的治疗作用

摘    要:目的:复制大鼠糖尿病肾病模型,研究复方丹参联合阿司匹林对大鼠糖尿病肾病的治疗作用及其机制。方法:选择体重180~200g的健康雄性SD大鼠64只,随机选取54只复制大鼠糖尿病肾病(DN)模型,其余大鼠作为正常对照(NC)组。实验第8周后,随机将模型组大鼠分为糖尿病肾病(DN)对照组、阿司匹林(DN+A)治疗组、复方丹参(DN+S)治疗组、复方丹参联合阿司匹林(DN+A+S)治疗组各10只。自模型成功之日起分别按相应方案治疗。检测空腹血糖(FBG)、血尿素氮(BUN)、血肌酐(Scr)、尿24h微量白蛋白(mALb)、肾组织丙二醛(MDA)、血栓素B2(TXB2),光镜观察大鼠肾组织形态学变化。比较治疗后各组之间上述指标的差异。结果:与DN对照组相比,DN+A+S组FBG、BUN、Scr、24hmAlb、MDA、TXB2均明显降低(P<0.05);DN+A组BUN、Scr、MDA、TXB2也明显降低(P<0.05),但FBG、24hmAlb无显著降低(P>0.05);DN+S组也见FBG、BUN、Scr、24hmAlb、MDA、TXB2明显降低(P<0.05)。DN+A+S组同DN+A组相比,上述指标变化更显著(P<0.05);同DN+S组相比,上述指标的改善同样更明显,P<0.05(FBG除外)。结论:复方丹参联合阿司匹林对大鼠DN的改善作用与其降低血糖、抗氧化应激损伤和改善肾脏微循环等有关。

Abstract:Objective:Through copying the model of rat diabetic nephropathy,this topic tells us the therapeutical effect and its mechanism of action of rat diabetic nephropathy by researching the koinonia of compound salvia associating with aspirin.Methods:Selecting 64 healthy male SD rats with the weight of 180~200g.And randomly selecting 54 ones among them,coped the model of rat diabetic nephropathy.The others be put on the list of normal control group.After 8 weeks,Randomly,the rats of group model is divided into 10 ones of diabetic nephropathy control group,10 ones of aspirin therapy group,10 ones of compound salvia therapy group and 10 ones of compound salvia associating with aspirin therapy group. Since the day of successfully copying the model of rat diabetic nephropathy,each group is given appropriate treatment The glucometer detects FBG. By diacetyl monoxime method,Blood urea nitrogen is detected.By picric acid test,Scr is detected.By RIA,urine 24hmALb are detected By TBAR, nephridial tissue MDA is detected.By ELISA.TXB2 are detected.Through light microscope,the morphological change of rat nephridial tissue is observed.And comparing the index difference among groups after treatment,all the results are analysed by Students test with SPSS14.0 for Windows software.Results:Comparing with DN control group,DN+A+S group FBG.BUN、Scr,24h urine mALb,MDA,TXB2 significantly lower,(P<0.05). DN+A group BUN、Scr、MDA、TXB2 reduction exists,(P<0.05);FBG,24h urine mALb exception(P>0.05).DN+S group FBG、BUN、Scr,24h urine mALb,MDA、TXB2 significantly lower,(P<0.05).Comparing DN+A+S group with DN+A control group,These indicators changes more significantry,P<0.05.Comparing DN+A+S group with DN+S control group,These indicators changes more significantly,(FBG exception)P<0.05. Conclusion:The first,three types of treatment are treatment of rats with the effect of DN,but compound salvia in combination with aspirin on the curative effect in rats significantly better than aspirin alone or the effect of compound salvia.The second,compound salvia in combination with aspirin for the improvement of the role of rats to reduce their blood sugar,anti-oxidative stress and improve the microcirculation of the kidney.
