摘    要:目的探讨雌激素受体2基因(ESR2)多态性与盐城地区青年胃腺癌的关联。方法应用聚合酶链反应-限制性片段长度多态性技术,对盐城地区100例青年胃腺癌患者(胃腺癌组)和100名青年胃溃疡患者(对照组)ESR2第5外显子RsaⅠ酶切多态和第8外显子AluⅠ酶切多态进行分析。结果(1)ESR2第8外显子AluⅠ酶切多态分析表明,胃腺癌组A等位基因频率明显高于对照组(P〈0.05,OR=1.975),胃腺癌组含等位基因A的Aa+AA基因型频率也明显高于对照组(P〈0.05,OR=2.144);(2)两组ESR2第5外显子RsaⅠ酶切多态基因型rr、Rr、RR频率和等位基因r、R频率的比较,差异均无统计学意义(P〉0.05)。结论ESR2第8外显子AluⅠ多态可能与盐城地区青年胃腺癌易感性有关,含等位基因A的Aa和AA基因型可能增加青年胃腺癌发病风险;而ESR2第5外显子RsaⅠ多态与盐城地区青年胃腺癌发病风险无关联。

Abstract:Objective To investigate the relationship of estrogen receptor 2 gene (ESR2) polymor-phism associated with gastric adenocarcinoma in Yancheng of China. Methods By polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method,the RsaⅠpolymorphism in exon 5 and the AluⅠpolymorphism in exon 8 of ESR2 were detected in 100 youths with gastric adenocarci-noma group and 100 youths with gastric ulcer(control group) in Yancheng. Results (1)The frequency of the allele A of AluⅠpolymorphism in exon 8 was significantly higher in gastric adenocarcinoma group than in the control group (P〈0.05,OR=1.975),so was the frequency of the Aa+AA geno-types (P〈0.05,OR=2.144). (2)The genotype distributions(rr,Rr and RR) and allele frequencies (r and R) of RsaⅠpoly-morphism in exon 5 were not significantly different between the two groups (P〉0.05). Conclusion The AluⅠpolymorphism in exon 8 of ESR2 may be associated with the susceptibility of gastric adenocarci-noma in Yancheng. The Aa+AA geno-type significantly elevated the risk suffering from the gastric ade-nocarcinoma. The RsaⅠpolymorphism in exon 5 of ESR2 is not associated with the risk for puffering from the gastric adenocarcinoma in Yancheng.
