补肾活血组方对大鼠卵巢颗粒细胞的作用

摘    要:目的探讨补肾活血组方对雷公藤致卵巢储备功能下降大鼠卵巢颗粒细胞凋亡及相关基因Bcl-2及Bax表达的影响。方法 12周龄SD雌性大鼠60只,随机分6组:补肾活血组方低、中、高剂量组,六味地黄药组,正常组,雷公藤组。观察大鼠卵巢组织病理形态、卵巢颗粒细胞凋亡相关因子Bcl-2及Bax的表达。结果补肾活血组方中、高剂量组卵巢组织的颗粒细胞层厚度和闭锁卵泡/生长卵泡的比值与雷公藤组比较,差异有统计学意义(P〈0.01或〈0.05)。补肾活血组方中、高剂量组的卵巢抑制凋亡基因Bcl-2表达增强、促凋亡基因Bax表达减弱,与雷公藤组比较,差异有统计学意义(P〈0.01或〈0.05)。结论补肾活血组方中、高剂量组能促进卵泡成熟,延缓颗粒细胞衰老,提高卵巢的储备功能。

Abstract:Objective To discuss the effects of Bushenhouxue formula on ovarian granulose cell apoptosis and related genes Bcl-2,Bax of SD rate by Leigongteng(GTW).Methods Sixty 12-week-old female SD rats were randomly divided into 6 groups(the Bushenhuoxue fromula low dose,middle dose,and high dose groups,the Liuweidihuang formula group,GTW group and the control group) and vagina shedding cell checking was done.The pathomorphological shape of ovarian tissue and expression of Bcl-2,Bax of ovarian granulose cell apoptosis was observed.Results The thickness of granular cell layers and the faslener/growth follicle ratio were significantly different in the Bushenhuoxue middle ana high dose groups compared with those in the GTW group(P 〈0.01 or 〈0.05).Bcl-2 was upregulated in the Bushenhuox-ue formula middle and high dose groups and Bax was downregulated compared with the GTW group(P〈 0.01 or〈 0.05).Conclusion The Bushenhuoxue middle and high dose formulae can prevent follicle premuture failure of granular cells and improve ovary reservation.
