人参皂苷Rg3对LoVo细胞迁移和侵袭影响

摘    要:目的 探讨人参皂苷Rg3对人结肠癌LoVo细胞迁移和侵袭能力的影响.方法 通过四甲基偶氮唑盐(MTT)比色法测定不同浓度Rg3对LoVo细胞增殖的影响,并确定用于研究抑制LoVo细胞迁移和转移功能作用的Rg3浓度和作用时间.设置空白对照组及Rg3实验组(5、10、20、30、40、50μmol/L),通过划痕试验,对LoVo细胞划痕加药处理24 h后,观察不同浓度Rg3对细胞迁移的影响.采用Transwell法观察不同浓度Rg3对细胞侵袭的影响.采用Western blot检测不同浓度Rg3对CD44、基质金属蛋白酶2(MMP2)、基质金属蛋白酶抑制剂2(TIMP2)、E-cadherin和β-catenin表达水平的影响.结果 MTT结果显示:0~30μmol/L Rg3处理LoVo细胞24 h对其生长无明显影响.随Rg3作用时间的延长和浓度的增加,Rg3对LoVo细胞抑制作用逐步增强.划痕试验和Transwell试验表明:Rg3浓度增加,对LoVo细胞迁移及侵袭能力抑制作用增强,浓度达15μmol/L以上时,Rg3对实验组有显著抑制作用.Western blot检测表明Rg3浓度的升高,伴随CD44、MMP2蛋白表达水平下降,TIMP2表达上升,但E-cadherin和β-catenin蛋白表达水平无明显改变.结论 人参皂苷Rg3对人结肠癌LoVo细胞迁移和转移能力有抑制作用,其抑制作用具有时间和浓度依赖性.

Abstract:Objective To investigate the effect of ginsenoside Rg3 on the migration and invasion of human colon cancer LoVo cells. Methods The proliferation of LoVo cells was determined by methyl thiazolyl tetrazolium ( MTT) method. The correlation of Rg3 concentration and intervention time was investigated. The control group and the experimental groups with 5, 10, 20, 30, 40 and 50μmol/L were established, and the scratch test was performed to observe the effect of Rg3 on cell migration at 24 h. Transwell method was used to evaluate cell invasion. Western blot was used to detect the expression of CD44, matrix metalloproteinase 2 ( MMP2) , tissue inhibitor of metalloproteinase 2 ( TIMP2) , E-cadherin and β-catenin. Results Treatment with 0-30 μmol/L of Rg3 for 24 h had no effect on the growth of LoVo cells. With the prolongation of Rg3 intervention time and increase of Rg3 concentration, inhibition of Rg3 on LoVo cells was also increased gradually. Rg3 inhibited LoVo cell migration and invasion in a dose-dependent manner.Meantime, Rg3 inhibited CD44 and MMP2 protein expression and increased TIMP2 expression, but did not affect the expression of E-cadherin and β-catenin protein. Conclusion Ginsenoside Rg3 has an inhibitory effect on LoVo cell migration and invasion in a time-and concentration-dependent manner.
