精眯对脾虚大鼠肠黏膜损伤的修复及葡萄糖吸收的影响

摘    要:目的 研究精眯对脾虚大鼠肠黏膜损伤的修复及葡萄糖吸收的影响.方法 将利血平造模成功的SD大鼠随机分为模型组,精眯低、中、高剂量组,阳性药表皮生长因子(EGF)组及健康SD大鼠作为正常组,每组6只,通过在体单向肠灌流检测对葡萄糖吸收的影响及制作病理切片观察肠黏膜损伤修复情况,利用实时定量PCR(RT-PCR)及免疫组化检测相关钠依赖性葡萄糖转运载体(SGLT-1)mRNA、易化性葡萄糖转运载体(GLUT-2)mRNA及SGLT-1、GLUT-2表达差异.结果 根据吸收速率常数Ka和有效渗透系数Papp结果显示,与正常组相比,模型组对葡萄糖的吸收显著减少;与模型组相比,精眯各给药组随剂量增加吸收增大;与正常组相比,模型组肠黏膜明显变薄受损,腺体数目减少,SGLT-1,SGLT-1 mRNA,GLUT-2,GLUT-2 mRNA明显下调;与模型组相比,给药组肠黏膜得到修复,SGLT-1、SGLT-1 mRNA、GLUT-2、GLUT-2 mRNA上调,其中以精眯高剂量组与EGF组最为显著.结论 精眯通过肠黏膜的修复促进葡萄糖的吸收.

Abstract:Objective To observe the effect of spermidine on the repair of intestinal mucosa injury and glucose absorption in rats with spleen deficiency. Methods The successful model SD rats induced by reserpine were randomly divided into five groups, including the model group, three different dosages of spermidine groups and the positive medicine EGF group. Meantime, the healthy SD rats were used as the normal group. Single-track intestinal perfusionin vivo was applied to detect the glucose uptake. Pathological section was used to observe the repair of intestinal mucosa. The changes of odium-dependent glucose transporter ( SGLT-1 ) and easy oxidation of glucose transporter ( GLUT-2 ) were detected by immunohistochemistry and RT-PCR. Results According to the absorption rate constant Ka and effective permeability Papp , the absorption of glucose in the model group was decreased in comparison with the normal group. However, the absorption of glucose in spermidine groups was increased in a dose-dependent mannerwhen compared with the model group. Treatment with spermidine obviously improved intestinal mucosa damage and the expression of SGLT-1 and GLUT-2 mRNA and protein in model rats, especially in high dosage of spermidine group. Conclusion Spermidine promotes the absorption of glucose by upregulation of the intestinal mucosa repair.
