大孔吸附树脂分离纯化泽泻中4种三萜类化合物的工艺研究

摘    要:目的研究大孔吸附树脂分离纯化泽泻中4种三萜类化合物的工艺。方法以泽泻中的24-乙酰泽泻醇A、泽泻醇B、23-乙酰泽泻醇B及11-去氧-23-乙酰泽泻醇B为考察指标,采用静态吸附和动态吸附2种方法,优选出对4种三萜类化合物的吸附性能最佳的大孔吸附树脂,并对其工艺进行筛选,确定了纯化泽泻4种三萜类化合物的最佳工艺参数。结果 D-4020型大孔吸附树脂用于同时分离纯化泽泻中4种三萜类化合物,其吸附洗脱性能良好。最佳工艺参数:上样液生药质量浓度为0.50 g/mL,D-4020型树脂与生药量质量比为1∶1(g∶g),分别用蒸馏水、50%(体积分数)乙醇各5 BV洗去杂质,然后用90%(体积分数)乙醇8 BV洗脱,洗脱流速为2 BV/h。收集90%乙醇洗脱液,烘干,所得固体样品中4种三萜类化合物的总质量分数为18.68%。结论在所确定的工艺条件下,D-4020型大孔吸附树脂用于同时富集泽泻4种三萜类化合物的效果最佳。

Abstract:Objective To study on the isolation and purification technology of four triterpenes from Alismatis Rhizoma by macroporous resin.Methods The optimization of the isolation was performed by static and dynamic adsorption parameters with 24-acetyl alisol A,alisol B,23-acetyl alisol B and 11-deoxy-23-acetyl alisol B in Alismatis Rhizoma as indicators.Results D-4020 macroporous resin showed the best performance for simultaneous separation and purification of 4 triterpenoids.Optimal parameters were as follows: the crude drug concentration of 0.50 g/mL,resin and the(g:g) with 1:1,first eluting with distilled water,then 50% ethanol eluting for 5 BV,and 90% ethanol for 8 BV with elution flow rate of 2 BV/h.90% ethanol eluting to collect and dry to obtain the solid samples with four triterpenoids’ content of 18.68%.Conclusion D-4020 macroporous resin in the process conditions may be suitable for the simultaneous purification of the triterpenoids.
