黄芩群体不同形态变异类型遗传多样性的AFLP分析

摘    要:目的研究黄芩群体形态变异类型的遗传多样性。方法采用扩增片段长度多态性(AFLP)技术,对人工栽培群体的8个形态变异类型黄芩进行多态性分析。结果通过筛选得到9对AFLP引物组合,共扩增出2 834个DNA条带,多态性条带为1332个,多态性位点平均为47.8%。聚类分析结果表明,8个形态变异类型黄芩划分为4类。其中,绿茎、小叶、少分枝和紫色花类型与普通类型各自聚为一类,绿茎、大叶、少分枝和淡紫色花类型与紫红茎、大叶、少分枝和紫色花类型聚为一类,绿茎、小叶、多分枝和淡紫色花类型,绿茎、大叶、多分枝和紫色花类型,绿茎、小叶、多分枝和淡紫色花类型与紫红茎、小叶、多分枝和紫色花类型聚为一类。结论人工栽培群体黄芩形态变异类型间存在较高多态性,遗传多样性较丰富。

Abstract:Objective To investigate the genetic diversity of morphological variation types in Scutellaria baicalensis Georgi.Method Using the AFLP technology,the polymorphisms of 8 cultivated variation types in S.baicalensis Georgi were analyzed.Results Using 9 pairs of AFLP primers screened,2 834 DNA bands were amplified,among which 1332 were polymorphic.The polymorphic locus was 47.8% in average.Cluster analysis indicated that 8 morphological variation types of S.baicalensis Georgi were grouped into 4 classes.The green stem,small leaf,few branch,purple flower type and the common type were classified into one class.The green stem,big leaf,few branch,mauve flower type and the purple stem,big leaf,few branch,purple flower type were classified into one class.The green stem,small leaf,multi-branch,mauve flower type,the green stem,big leaf,multi-branch,purple flower type,the green stem,small leaf,multi-branch,mauve flower type,and the purple stem, small leaf,multi-branch,purple flower type were classified into another class.Conclusion The artificially cultivated S.baicalensis Georgi has high polymorphism among morphological variation types and rich genetic diversity.
