摘    要:目的通过离体实验观察人肠道菌群对芦丁代谢的情况。方法将人肠道内细菌加入GAM培养基,37℃离体厌氧培养24 h得到肠道菌孵育液,对照组取经灭活的肠道菌孵育液与芦丁37℃厌氧温孵,代谢组取有活性的肠道菌孵育液与芦丁37℃厌氧温孵,采用TLC法和HPLC法对芦丁的肠菌代谢产物进行分析。结果对照组24 h内芦丁水解34.2%;代谢组24 h内芦丁减少98.2%,并最终代谢完全。对照组在48 h内一直有槲皮素出现;代谢组有槲皮素出现,36 h时消失,15~21 h时代谢组的槲皮素含量升高,且明显高于对照组。结论离体培养的人肠道菌群可以对芦丁进行代谢,可能的代谢途径有2种:一种途径是糖苷键断裂转化为槲皮素,槲皮素经代谢转化为其他成分;另一种是不经槲皮素直接转化为其他物质。

Abstract:Objective To study on the metabolism of rutin by human intestinal bacteria in vitro.Methods Human intestinal bacteria was cultured in the GAM medium in vitro under the anaerobic environment at 37 ℃ condition.Rutin was put into the incubated buffer with inactivated human intestinal bacteria in control group,and with active bacteria in metabolic group.TLC and HPLC were used to analyze rutin and its metabolites.Results In control group,the content of rutin decreased by 34.2% gradually within 24 h,however,in metabolic group,it decreased by 98.2% gradually within 24 h and all rutin was metabolized within 48 h.Quercetin was found in control group within 48 h and it was also found in metabolic group before 36 h.The content of quercetin was significantly higher than control between 15 h and 21 h.Conclusion Rutin was metabolized by intestinal bacteria in vitro,and there may be two possible pathways: rutin was transformated into quercetin through breaking the glycoside bond or other substances independent of quercetin.
