山茱萸环烯醚萜总苷对实验性糖尿病肾病变的保护作用

摘    要:目的 研究山茱萸环烯醚萜总苷 (HXMTZG)对链脲佐菌素 (STZ)诱导的大鼠糖尿病肾病变 (DN)的保护作用。方法大鼠ipSTZ 6 0mg/kg ,1周后血糖值高于 16 .7mol/L者则纳为糖尿病大鼠。将糖尿病大鼠随机分为模型组、氨基胍 (AG)组 (0 .1g/kg ,ig)和HXMTZG组 (0 .5、1.0g/kg ,ig) ,同期另设正常组 ,各组连续灌胃给药 12周。实验结束后测定血糖和尿微量白蛋白 (UMA) ,取肾脏经HE和PAS糖原染色后进行光镜检查。结果 模型组大鼠血糖、UMA排出量均高于正常组 ,HXMTZG及AG对血糖基本无降低作用 ,但两者均能显著降低UMA的排出 (P <0 .0 1) ;光镜检查显示模型组呈现早 -中期糖尿病肾病变 ,而HXMTZG及AG组均能不同程度地减轻肾小球系膜增生、GBM增厚、糖原沉积及AE透明变等。结论HXMTZG能延缓实验性DN大鼠的发生与发展 ,对临床治疗DN具有积极的意义

Abstract:OBJECTIVE To investigate into the protecting effect of iridoid glycoside in Fructus Corni Officinalis (HXMTZG) on rat's diabetic nephropathy induced by streptozotocin (STZ). METHOD Rats were given ip STZ 60 mg/kg and those with the blood sugar level higher than 16.7 mol/L were considered as rats with diabetes. These rats were divided randomly into a model group, an aminoguanidin (AG,China) group (0.1 mg/kg, ig,China) and a HXMTZG group (0.5, 1.0 mg/kg, ig). A normal group was formed. Each group of rats were given the medicine for 12 weeks before blood sugar and urine trace albumin were determined and light microscope was used in the experiment. RESULT The output of blood sugar and albumin of the model group were both higher than those of the normal group, and HXMTZG and AG had no marked effect in reducing blood sugar but could reduce UMA output. Light microscope detected that diabetic nephropathy occurred in the model group, and glomerular mesangium hyperplasia, thickening of GBM, glycogen deposition and AE hyaline degeneration were relieved in HXMTZG and AG groups. CONCLUSION HXMTZG can delay the occurrence of nephropathy of experimental rats.
