白藜芦醇对裸鼠胃癌移植瘤中PDCD5蛋白表达及细胞凋亡的影响

摘    要:目的探讨白藜芦醇(Res)对裸鼠胃癌移植瘤细胞凋亡相关基因PDCD5蛋白表达及细胞凋亡的影响。方法将BGC-823细胞复苏后,通过右前肢腋部皮下移植构建裸鼠胃癌移植瘤模型。建模成功后的荷瘤裸鼠随机分为空白对照组、Res组、顺铂组和联合用药组(白藜芦醇+顺铂),各组分别在成瘤部位皮下注射0.2ml相应的药物,每日1次,治疗1周。停药后将裸鼠处死,剥离肿瘤组织。采用免疫组织化学方法检测移植瘤组织中PDCD5蛋白表达,采用原位末端标记法(TUNEL)检测移植瘤组织中细胞凋亡情况。结果 Res组中PDCD5蛋白表达水平明显高于空白对照组,但低于顺铂组和联合用药组,上述各组两两相比,差异均有统计学意义(P<0.05)。TUNEL法检测到胃癌移植瘤中凋亡细胞核呈棕褐色,染色深浅不一,染色质分布不均,空白对照组中仅见到少数凋亡细胞,Res组、顺铂组和联合用药组凋亡细胞数依次增加,上述各组两两相比,差异均有统计学意义(P<0.05)。结论白藜芦醇可上调裸鼠胃癌移植瘤细胞中PDCD5的蛋白表达,表明其可能通过上调PDCD5的表达进而诱导细胞凋亡。

Effects of resveratrol on protein expression of PDCD5 and apoptosis in gastric carcinoma transplanted tumor carried by nude mice

Abstract:Objective To explore the effects of resveratrol(Res) on the protein expression of apoptosis related gene PDCD5 and apoptosis in gastric carcinoma carried by nude mice.Methods The anabiotic BGC 823 cells were injected under the right axilla skin in nude mice to establish transplanted tumor model.After the model was established,the carrying transplanted tumor nude mice were randomly divided into blank control group,Res group,cisplatin group and combination group (resveratrol + cisplatin),and each group was given subcutaneous injection of 0.2ml corresponding drugs everyday for a week.Then,the mice was executed and the tumor tissues was collected.Immunohistochemistry SP and TUNEL methods were used to examine PDCD5 protein expression and the cell apoptosis of transplanted tumor of human gastric adenocarcinoma respectively.Results The protein expression of PDCD5 protein was significant higher in Res group than that in blank control group(P<0.05),while it was lower than that in cisplatin group and combination treatment group(P<0.05).The apoptotic cells showed nuclei brown,chromatin distribute unevenly and dyeing depth varies detected by TUNEL method.There was little apoptotic cells in the blank control groups,and in experiment groups the number of apoptotic cells increased,while the Res group was less than that in the cisplatin group,and combination treatment group had the most poptotic cells(P<0.05). Conclusion The resveratrol could up-regulated the expression of PDCD5 protein in transplanted tumor of gastric adenocarcinoma, it indicates that the resveratrol may induce apoptosis through up-regulating the expression of PDCD5.
