血清microRNA-21在食管鳞状细胞癌中表达的临床意义

目的 探讨食管鳞状细胞癌（ESCC）患者血清中外核体microRNA-21（miRNA-21）表达水平的临床意义.方法 从51例ESCC患者（ESCC组）血清中分离出外核体,提取总RNA,通过实时定量PCR技术检测miRNA-21的表达情况.以良性病变且无全身性炎性反应的41例患者作为对照组.结果 ESCC组患者血清中外核体miRNA-21表达水平明显高于对照组患者（P〈0.01）,而在已去除外核体的剩余血清中检测不到miRNA-21.ESCC患者血清外核体能在体外诱导食管鳞状细胞癌细胞增殖.外核体miRNA-21高表达亚组中年龄≥68岁患者比例、T3/T4型比例、III/IV期患者比例明显高于低表达亚组,而发生癌症转移患者的比例明显降低（P〈0.05,P〈0.01）.结论 ESCC患者血清中外核体miRNA-21表达明显上调,外核体miRNA-21表达水平与肿瘤进展和侵袭性密切相关,提示外核体miRNA-21可能是肿瘤治疗的一个有效靶标.

Clinical significance of serum microRNA-21 in human esophageal squamous cell carcinoma

Objective To investigate the clinical significance of the detection of microRNA-21（miRNA-21） expression in the serum exosomes of the patients with esophageal squamous cell carcinoma（ESCC）.Methods miRNA 21 was separated from the serum exosomes in 51 patients with ESCC and its expression was detected using real time quantitative PCR expression.Serum from the 41 patients with benign lesions but no systemic inflammation was collected as control group.Results Serum exosome from the ESCC patients could induce esophageal squamous cell carcinoma cell proliferation in vitro.The expression of miRNA-21 in serum exosomes of ESCC was significantly higher than that in the patients with benign lesions（P 0.01）.In the remaining serum that the exosomes were removed,miRNA-21 could not be detected.The expression of miRNA-21 has relationship with tumor classification,lymph node positive,cancer metastasis and/or clinical typing.Conclusion Compared with the control group,the serum exosomes miRNA-21 was significantly up regulated in the patients with ESCC,which is closely related to the progression and invasion of tumor,suggesting that exosome miRNA-21 may be an effective target for tumor therapy.
