逍遥散对抑郁症模型大鼠脑和结肠中五羟色胺表达的影响

摘    要:目的观察中药逍遥散对抑郁模型大鼠脑和结肠中五羟色胺(5-HT)表达的影响。方法 40只雄性SD大鼠随机分为正常组、模型组、氟西汀组、逍遥散组各10只,后3组采用慢性应激方法结合孤养法制备抑郁大鼠模型,正常组及模型组给予生理盐水,氟西汀组给予2 mg/ml的氟西汀,逍遥散组给予1.25g/ml的逍遥散,均2 ml,连续21 d。观察各组体质量增长、自主活动、糖水消耗的变化,并用免疫组化SABC法检测各组大鼠右侧大脑皮质和结肠中5-HT的表达。结果与正常组比较,模型组体质量增长数、主动活动次数及糖水消耗量下降,脑皮质及结肠中5-HT的MD值及YP值均均降低(P<0.01)。与模型组比较,氟西汀组及逍遥散组各项指标均有改善,且逍遥散组改善优于氟西汀组(P<0.05,P<0.01)。结论逍遥散的抗抑郁作用可能与增加脑内及结肠中单胺神经递质5-HT含量有关。

Effect of Xiaoyaosan on the txpression of 5-HT in the brains and colons of depression rats model

Abstract:Objective To investigate the effect of Xiaoyaosan on serotonin(5-HT) expression in the brains and colons of depression model rats.Methods Forty SD male rats were randomly divided into 4 groups:normal group,model group,the fluoxetine group,and Xiaoyaosan treatment group.The rats were subjected to chronic unpredicted stress and separation to establish depression model rats.The autonomic activities and sugar consumption of the rats,behavior were compared and the 5-HT expression in the right cortex and in the colon of the rats in each group were examined by using immunohistochemical SABC method.Results Compared with the control group,Xiaoyaosan treatment group can count significantly increased weight gain(P 0.05),significantly increased the autonomic activities and sugar water consumption in model rats(P 0.05),pituitary and colon of right rats hippocampus 5- HT increased significantly(P 0.01),compared with model group,all parameters in fluoxetine group and Xiaoyaosan treatment group were improved significandy,and there were difference between fluoxetine group and Xiaoyaosan treatment group(P 0.05,P 0.01).Conclusion The Xiaoyaosan's antidepressant effect may be associated with an increase in brain and colon monoamine neurotransmitters 5-HT content.
