缺氧大鼠血管内皮功能的变化及通心络超微粉的干预作用

摘    要:目的观察缺氧大鼠血管内皮功能的变化及通心络超微粉的干预作用。方法50只大鼠随机分为空白对照组、缺氧模型组、通心络大剂量组、通心络中剂量组和通心络小剂量组。缺氧模型组与通心络组大鼠灌胃后置于常压低氧舱内,每天7h,每周6d,共计5周。实验结束后检测大鼠动脉血气分析,HE染色观察大鼠胸主动脉的形态学改变,透射电镜观察主动脉内皮细胞超微结构的改变,放射免疫均相竞争法检测大鼠血浆内皮素(ET)含量,硝酸酶还原法检测大鼠血清一氧化氮(NO)含量,双抗夹心ELISA法测定大鼠血浆血管性假血友病因子(vWF)的含量。结果与空白对照组比较,缺氧模型组大鼠动脉血PaO2显著降低(P<0.05);血浆ET-1与血清vWF水平显著升高,血清NO水平显著下降,差异均有统计学意义(P<0.01);通心络超微粉可明显升高血氧分压(P<0.05),降低血浆ET-1与血清vWF水平(P<0.05),升高血清NO水平(P<0.01)。光镜下可见缺氧模型组大鼠胸主动脉结构改变明显,透射电镜下主动脉内皮细胞超微结构明显改变,而通心络组病变减轻。结论缺氧可造成大鼠血管内皮结构破坏和功能损伤,通心络超微粉对缺氧引起的血管内皮结构破坏和功能损伤具有明显改善作用。

The effect of hypoxia on the function of vascular endothelial and intervention of Tongxinluo capsule ultrafine

Abstract:Objective To investigate the vascular endothelial function and interventional effect of Tongxinluo capsule ultrafine in hypoxia rats model. Methods Fifty male Wistar rats were randomly assigned to following groups : control group,hypoxia model group, TXL high-dose group, TXL middle-dose group and TXL low-dose group. The rats of hypoxia model group and TXL group were underwent normal pressure hypoxia cabin, 7 hours every day and 6 days every weeks, the experimental period was add up to 5 weeks. In the end of experiment, arterial blood was detected by arterial blood gas analy sis instrument, the morphology of thoracic aorta was observed according to HE staining and aortic endothelial cells ultrastructure according to TEM,the content of ET in plasma was measured withradio-immunity method, the content of NO in serum was measured with nitrate-reduction method, the content of vWF in plasma was measured with enzyme linked immunosorbent assay (ELISA). Results Compared with control group, arterial partial pressure of oxygen was lower obviously( P 〈 0.05), the content of ET and vWF in plasma significantly increased, the content of NO in the serum significantly decreased in hypoxia model group( P 〈 0. 01 ). The Tongxinluo capsule ultrafine could increase the arterial partial pressure of oxygen of model rats( P 〈 0.05) ,decreased the levels of ET and vWF in plasma( P 〈 0.05) ,raise the levels of NO in serum( P 〈 0.01 ). The structure of the thoracic aorta and aortic endothelial cells ultrastructure were significantly changed in the hypoxia model group in LM and TEM. And the lesion was lightened in the TXL groups. Condusion Hypoxia could cause structural damage and function injury of vascular endothelin in the rats model, Tongxinluo capsule on hypoxia-induced vascular endothelial damage and dysfunction with a marked improvement in the role.
