抑癌基因PTEN与人错配修复基因hMSH2在宫颈癌变过程中的表达及意义

摘    要:目的探讨抑癌基因PTEN与人错配修复基因hMSH2在宫颈癌中的表达及其与宫颈癌发生的关系。方法应用免疫组织化学方法检测34例正常宫颈、101例宫颈上皮内瘤样病变(CINI、CINII、CINIII)、46例宫颈癌等不同组织中PTEN与hMSH2的表达情况。结果PTEN蛋白在正常宫颈、CIN和宫颈癌各组组织中阳性表达率逐渐降低,其中宫颈癌组与其余各组比较差异均有统计学意义(P<0.01);CIN组与正常宫颈组间比较差异无统计学意义(P>0.05),CINIII组与CINI组比较,差异有统计学意义(P<0.05)。hMSH2在正常宫颈、CIN和宫颈癌各组组织中阳性表达逐渐增高,宫颈癌组与其余各组比较差异有统计学意义(P<0.01),CIN组与正常宫颈组比较,差异亦有统计学意义(P<0.01);CINIII组与CINI组比较差异有统计学意义(P<0.05)。宫颈癌组织中PTEN和hMSH2蛋白表达经Spearman相关分析,显示呈负相关关系(P<0.05)。结论PTEN和hMSH2在宫颈癌发生发展中发挥重要作用,联合检查可以作为宫颈癌筛查、早期诊断及基因治疗的指标之一。

The expression and significance of PTEN and hMSH2 in the development of cervical carcinoma

Abstract:Objective To explore the expression of tumor suppressor gene PTEN and human mismatch repair gene hMSH2 in cervical carcinoma and the relationship between them. Methods Immunohistochemistry was applied to detect the expression of PTEN and hMSH2 in normal cervical tissues (34 cases), cervical intraepithelial neoplasia (CINⅠ, CINⅡ, CIN Ⅲ, 101 cases) and cervical carcinoma (46 cases). Results The expression of PTEN protein decreased gradually in normal cervical tissues, CIN and cervical carcinoma. The expression of PFEN protein in cervical carcinoma group was significantly lower than that in other groups ( P 〈 0.01 ). Compared between CIN and normal cervical tissue, there was no significant difference( P 〉 0. 05). The expression of PTEN protein in CINⅢ was significantly lower than CINI( P 〈 0.05). The expression of hMSH2 protein increased in normal cervical tissues, CIN and cervical carcinoma group gradually, compared with other groups, the expression of hMSH2 protein in cervical carcinoma group was significantly higher( P 〈 0. 01), compared between CIN and normal cervical group there were significant differences( P 〈 0.01 ), the expression in CINⅡ group was hig her than CINⅠ group( P 〈 0.05). Based on Speaman correlation analysis, there were negative correlation in cervical carcinoma, both the expression of PFEN and hMSH2 protein( P 〈 0. 05). Conclusion PTEN and hMSH2 protein may play important roles in the development of cervical carcinoma. It may be helpful for predicting and diagnosing cervical carcinoma in early stage.
