三氧化二砷与人参皂甙Rg3联合对人肝癌SMMC-7721细胞侵袭转移能力的影响

摘    要:目的探讨三氧化二砷(As2O3)和人参皂甙Rg3(ginsenoside Rg3)联合运用对人肝癌SMMC-7721细胞黏附侵袭转移能力的影响以及与单独运用的差异。方法采用MTT法观察经As2O3、人参皂甙Rg3单独与联合作用后人肝癌SMMC-7721细胞对基底膜黏附能力的影响;Transwell膜侵袭系统观察As2O3与人参皂甙Rg3单独与联合作用后SMMC-7721细胞游走与穿透基底膜能力的影响。结果As2O3和人参皂甙Rg3单独与联合应用均能显著抑制人肝癌SMMC-7721细胞与Marigel的黏附,抑制细胞游走与穿透基底膜的能力(P<0.05),其联合抑制作用优于单独运用(P<0.05)。结论As2O3和人参皂甙Rg3均有抗肿瘤侵袭转移作用,两者联合可增强抑制效应。

Abstract:Objective To study the difference of arsenic trioxide (As_2O_3) or ginsenoside Rg3 alone,as well as their combination application in adhesion and invasion metastasis of SMMC-7721 human liver cancer cell in vitro.Methods MTT was used to study the effect of adhesion treated by As_2O_3 combined with ginsenoside Rg3 on SMMC-7721 cell.The Transwell Chamber membrane invasion culture system was used to study the invasion and metastasis.Results The As_2O_3 or ginsenoside Rg3 alone and their combination application could significantly inhibit SMMC-7721 cell adhesion with Matrigel,movement and migration through Matrigel than those in the control group (P< 0.05).Their combination application enhanced the effect compared with As_2O_3 or ginsenoside Rg3 alone (P< 0.05).Conclusion As_2O_3 or ginsesnoside Rg3 alone have the effect of antagonizing hepatocarcinoma cell adhesion and invasion metastasis,their combination application could enhance the effect.
