Abstract:Objective To explore the relationship between the expression of microRNA1395p(miR-139-5p)and cadherin-related protein(CTNNB1)in non-small cell lung cancer tissues and the radiosensitivity of patients.Methods One hundred and fifty-six patients with non-small cell lung cancer who were treated in the Seventh Department of Radiochemistry and Oncology of Tangshan People's Hospital from February 2017 to May 2021 were selected as the research objects(research group).In addition,86 patients with non-small cell lung cancer patients with normal lung tissue adjacent to cancer were selected as the control group.According to the radiotherapy sensitivity evaluation criteria,patients in the study group were divided into progression subgroup(n=28),stable subgroup(n=42),and remission subgroup(n=86).Real-time fluorescence quantitative PCR and immunohistochemistry were used to detect the expression of miR-139-5p and CTNNB1 protein in lung cancer tissue and normal lung tissue adjacent to cancer.The relationship between the expression of miR-139-5p and CTNNB1 protein in cancer tissues of patients with non-small cell lung cancer and the clinicopathological characteristics and radiosensitivity were analyzed.Spearman method was used to analyze the correlation between the expression of miR-139-5p and the positive expression of CTNNB1 protein in cancer tissues of patients with non-small cell lung cancer.Logistic regression analysis of factors affecting radiosensitivity of patients with non-small cell lung cancer.Results The expression level of miR-139-5p in cancer tissues of the study group was lower than that of the control group(χ2/P=15.204/0.000),and the positive expression rate of CTNNB1 protein was higher than that of the control group(χ2/P=59.206/0.000).The proportion of patients with low expression of miR-139-5p with lymph node metastasis,low tissue differentiation,and TNM stage IV was higher than that of patients with high expression of miR-139-5p(χ2/P=16.006/0.000,24.669/0.000,22.213/0.000).The proportion of patients with negative expression of CTNNB1 protein with lymph node metastasis,low tissue differentiation,and TNM stage IV was lower than that of patients with positive expression of CTNNB1 protein(χ2/P=21.922/0.000,22.959/0.000,30.108/0.000).The expression level of miR-139-5p in cancer tissues of patients with non-small cell lung cancer was negatively correlated with the positive expression of CTNNB1 protein(r/P=-0.503/0.000).The low expression rate of miR-139-5p and the positive expression rate of CTNNB1 protein in the remission subgroup,stable subgroup,and progress subgroup increased sequentially(χ2/P=25.762/0.000,18.721/0.000).Positive expression of CTNNB1 protein and high TNM stage are independent risk factors affecting radiosensitivity[OR(95%CI)=2.872(1.546-5.335),2.732(1.459-5.115)],high expression of miR-139-5p affects radiosensitivity Protective factors[OR(95%CI)=0.845(0.750-0.952)].Conclusion In non-small cell lung cancer tissues,the expression of miR-139-5p is down-regulated,and the positive expression of CTNNB1 protein is up-regulated,which affects radiosensitivity to a certain extent.
