通心络制剂对大鼠脑梗死灶周突触和星形胶质细胞的影响

摘    要:目的研究通心络对大鼠脑梗死周边区星形胶质细胞、突触和运动功能的影响。方法将120只大鼠大脑中动脉闭塞脑梗死模型随机分为通心络组、抑制组、盐水组和对照组,以横木行走实验评定运动功能,在5、10、15d应用电镜和免疫组织化学方法观察大鼠脑梗死周边区星形胶质细胞、突触数及胶质纤维酸性蛋白和突触素等的变化。结果通心络组运动功能评分10d、15d分别为(3.2±0.3)和(5.8±0.9)分,高于对照组、抑制组(P<0.01);电镜观察,10d、15d脑梗死周边区星形胶质细胞分别为(5.7±0.3)、(8.4±0.7),突触数目分别为(11.9±1.1)、(15.9±1.3)(个/8000×HP),两者均明显多于对照组和抑制组(P<0.01);免疫组化染色观察,脑梗死周边区胶质纤维酸性蛋白和突触素阳性信号密度值也在10d、15d高于对照组和抑制组(P<0.01)。结论通心络治疗可使脑梗死大鼠星形胶质细胞增殖,加强突触重建和功能修复,促进运动功能恢复。

Effect of Tongxinluo on astrocyte and synapse around cerebral infarction lesion in rats

Abstract:Objective To investigate the effect of traditional Chinese medicine Tortgxinluo on astrocytes, synapses and motor function in the area around cerebral infarction (area A) in rats. Methods 120 rats were randomly divided into 4 groups, Tongxinluo treatment group, inhabitation group, saline group and control group. The :motor function was examined via beam walking test. The changes were observed included astrocytes, synapses, the glial fibrillary acidic protein (GFAP) and synaptophysin in area A at 5, 10 and 15d after the model was set up. Results The motor function mark in the Tongxinluo treatment group was (3.2 ± 0.3)in 10d and (5.8±0.9) in 15d, respectively, significantly higher than that in control or inhabitation group ( P 〈 0.01 ). With electron microscope in Tongxinluo treatment group, the astrocytes was (5.7 ±0.3) and (8.4 ±0.7) in 10 and 15 days, and synapses was ( 11.9 ±1. 1) and ( 15.9 ±1.3) in 10 and 15 days, which were significantly higher than those in control or inhabitation group ( P 〈 0.01 ). With immunohistochemistry, it showed that the optical density of GFAP and synaptophysin in the Tongxinluo treatment group were significantly higher than that of control or inhabitation group in area A at 10 and 15 days ( P 〈 0.01 ). Conclusion It demonstrated that Tongxinluo could induce astrocyte proliferation, augment realignment and promote the motor function recovery.
