氨利酮诱发V79细胞HGPRT位点突变研究

摘    要:本文观察了国产心血管疾病新药氨利酮在V79细胞HGPRT位点正向突变试验系统中的诱变作用。结果表明:氨利酮在S9情况下,各浓度组突变体频率与对照组相比均未达到MF≥3SMF的标准,也无剂量反应关系;但相同浓度在+S9情况下,除高浓度组外,蓁三组MF随浓度增加而升高,且次高浓度组MF是对照组的3.8倍,故在本试验条件下氨利酮是需活化的诱变剂。

Abstract:Inducibility of 6-thioguanine-resistant mutants by a new medicine-amrinone was investigated. In the absence of S9. no mutagenic effect was found. In the presence of S9, mutant frequencies were increased with am-rinone concentration from 10uM to 100uM, but decreased at 200uM. At 100uM mutant frequency was increased more than three-fold over spontaneous mutant values. Thus it was suggested that amrinone can induce mammalian cells gene mutation.
