摘    要:目的探讨LIGHT对人食管癌细胞的体外抑制效应的可能机制.方法以DOTAP脂质体介导LIGHT-Fc基因转染人食管癌细胞株Eca109,通过免疫组织化学及流式细胞仪检测来观察LIGHT-Fc转染对Eca109细胞上ICAM-1表达水平的影响.结果 Eca109细胞表达LIGHT-Fc基因上调了细胞表面ICAM-1的表达水平.结论 LIGHT-Fc基因转染上调人食管癌细胞株上ICAM-1的表达可能是其体外抑瘤效应的一种机制,但全面评价有待进一步的研究.

Abstract:Objective To investigate the possible mechanisms for suppression effect of LIGHT on human esophageal carcinoma cell in vitro. Methods LIGHT-Fc expression vector was transfected into human esophageal squamous carcinoma cell line, Eca109 by DOTAP liposomal. The influence of LIGHT-Fc gene transfection to the expression of intercellular adhesion molecule-1(ICAM-1) was analysised by immunohistochemistry and FACS. Results The data shows that LIGHT-Fc gene transfection upregulates the expression of ICAM-1 on Eca109. Conclusion The upregulation of ICAM-1 may be a sort of mechanisms for the suppression effect of LIGHT on Eca109 in vitro, but the extensive evaluation still needs careful study.
