洛伐他汀对肾小球系膜细胞增殖及细胞周期时相的影响

目的探讨洛伐他汀对人肾小球系膜细胞（HMC）增殖和细胞周期时相的影响及机制。方法用３H-TdR掺入量法测定HMC增殖水平,以流式细胞术测定HMC的细胞周期。结果洛伐他汀剂量依赖性抑制HMC增殖,从细胞周期水平,洛伐他汀可使HMC由G１期向S期的进展受阻,这些抑制作用可被外源甲羟戊酸和法呢醇恢复。结论洛伐他汀是HMC增殖的抑制剂,本研究为重新认识和评价他汀类药物在治疗肾脏疾病中的作用提供了一定的理论及实验依据。

Inhibitory effects of lovastatin on the proliferation and cell cycle phase of cultured human glomerular mesangial cells in vitro

Objective To study the effects and mechanism of lovastatin on cell cycle phase and prolifera-tion of cultured human glomerular mesangial cells in vitro.Methods HMC proliferation was determ-ined by 3H-Thymidine incorporation.HMC cell cycle was measured by flow cytometric analysis Results Lova statin was found to inhibit HMC proliferation in a dose-dependent manner.Flow cytometric analysis demonstrated that lovstatin induced G1/S transition arrest.Concomitant addition of mevalonate or farnesol restored all the inhibitory effect of lovstatin on HMC.Conclusion Lovastatin is a HMC proliferation inhibitor.It provides an experimental evidence for re-evaluate renal protective effect of HRI,which has already been widely used in clinical treatment.
