中国妇女维生素D受体基因型与骨密度及骨钙素关系的研究

摘    要:为探讨中国妇女维生素D受体(VDR)基因型与骨密度(BMD)及含钙素间的关系,笔者采用聚合酶链反应、限制性酶切技术和Southern杂交技术,对202名北京地区汉族妇女的VDR基因型进行了分析。另外,采用双能X线法测量BMD,并采用放免法测定骨钙素。采用协方差分析的方法对结果进行分析处理。结果发现:中国妇女VDR基因型分布显著不同于高加索人种;绝经后妇女中bb与aa基因型在股骨颈及大转子区呈对应较低的BMD值;未发现VDR基因型与骨钙素间相关。因此,中国妇女VDR基因型与BMD间有一定关联,但其对骨质疏松发病的预测价值还有待深入研究。

Abstract:Objective To investigate the correlations between vitamin D receptor (VDR) gene polymorphisms, and bone mineral density(BMD) as well as osteocalcin. Methods We usedPCR-RFLP and Southern hybridization analysis to determine VDR genotypes in 202 Chinese women. BMD was measured by dual-energy X-ray absorptiometry (DEXA) and serum osteocalcin level was determined by radioimmunoassay. Analysis of covariance was used to test differences in mean BMDs across genotypes. Results We realized that VDR genotype frequencies in Chinese women were apparently different from those of Caucasians. In group of postmenopausal women, "bb"or"aa" genotype was related to lower BMD in Femoral Neck and Trochanter. No association was found between VDR genotypes and the serum osteocalcin level. Conclusion There was some association between VDR genotypes and BMD in Chinese women. lts function in predicting osteoporosis still requires further study.
