摘    要:目的 探讨大鼠脑组织缺血后不同时间脑红蛋白(Ngb)mRNA表达及法医学意义。方法 建立Wistar大鼠脑组织缺血模型,应用一步法逆转录-聚合酶链式反应(one step RT-PCR)技术与计算机图像分析技术检测脑组织缺血后不同时间Ngb mRNA表达的差别。结果 脑缺血后Ngb mRNA表达迅速升高,缺血1min达高峰,缺血5min表达迅速降低,但仍高于假手术组水平,10min后随缺血时间延长表达再度逐渐增强。结论大鼠脑组织缺血后Ngb mRNA的表达呈现时相性的变化,可在法医实践中用于早期脑缺血时间的推断。

Abstract:Objective To find out the value of Ngb mRNA in estimating the trauma time in forensic medicine aspect by observing expression of Ngb mRNA in different time after cerebral ischemia. Methods A rat model of cerebral ischemia was established. The expression of Ngb mRNA in different time after cerebral ischemia was detected by one step RT-PCR method and computer image analysis system. Results The expression of Ngb mRNA rose quickly after cerebral ischemia, reached peak value after 1 min. After 5 min it decreased quickly, but obviously higher than that in the sham operated group. After 10 min it rose gradually again with the prolongation of ischemia time. Conclusion The expression of Ngb mRNA changes time-dependently after cerebral ischemia, and it may be used to estimate the early time of ischemia trauma in forensic practice.
