围绝经期综合征实验动物模型的比较分析

摘    要:目的 比较围绝经期综合征的两种实验动物模型,为围绝经期综合征的实验研究奠定良好的基础,从而为治疗该综合征提供理论依据。方法经阴道脱落细胞涂片检查发情期没有规律改变的15月龄雌性大鼠6只作为围绝经期组,取3月龄雌性SD大鼠12只,6只设为青年空白对照,6只从背部行去势手术,术后半月行阴道脱落细胞涂片检查发情期没有规律改变,设为青年去势组,采用放免法(RIA)检测血清雌激素(场)、卵泡刺激素(FSH)和黄体生成素(LH)水平,比较各组大鼠血清性激素水平的变化。结果围绝经期组及青年去势组Fa,FSH,LH水平均显著低于青年对照组(P〈0.05);青年去势组与围绝经期组相比E2水平显著下降(P〈0.05),FSH,LH水平差异无统计学意义(P〉0.05)。结论去势模型与15月龄围绝经期雌鼠均可以模拟女性围绝经期激素水平的改变,但两种方法相比,15月龄围绝经期雌鼠激素变化更接近事实,用此模型研究围绝经期综合征理论上优于去势模型。

Abstract:Objective To compare two kinds of animal models of climacteric syndrome and to provide a theoretical basis for experimental research and treatment of the syndrome. Methods Fifteen-month female rats without regular oestrum were chosen as climacteric syndrome group and 3-month female rats were divided into two groups(control group and ovariectomy group).The rats in ovariectomy group were ovariectomized and had no regular oestrum at day 15 after operation.Levels of serum E_2,FSH and LH were determined by RIA. Results Compared with the control group, the levels of these hormones were significantly lower in the other groups.Compared with climacteric group,the level of E_2 in ovariectomy group decresed significantly,but there were no significant changes of FSH,LH. Conclusion The changes of sexual hormones in climacteric occurred in both animal models.Comparing two kinds of animal models,the changes of climacteric group are more consistent with the reality.It is better than ovariectomy model in terms of theory and reality.
