摘    要:目的探讨白及无菌苗增殖的影响因子以及活性炭对无菌苗生长的影响。方法利用TDZ、NAA和肌醇3因素3水平进行L9(34)正交实验设计,以增殖倍数为指标,研究白及无菌苗增殖的影响因子。结果 TDZ和肌醇对无菌苗增殖具有显著影响,NAA无显著影响。诱导无菌苗增殖的最适宜组合是Fast培养基+TDZ 5.0 mg/L+NAA 0.05 mg/L+肌醇50 mg/L,无菌苗增殖倍数达到了7.3倍。适宜无菌苗生长的活性炭添加量为0.05 g/L。结论不同因素对白及无菌苗增殖的影响有显著差别,影响最大的是TDZ和肌醇,NAA不影响白及无菌苗的增殖。最佳组合适于白及成苗及增殖,繁殖系数高,无菌苗生长健壮,可应用于白及的组培快繁。

Abstract:Objective To explore the factors affecting plantlets proliferation in vitro of Bletilla striata, and the effects of activated charcoal on plantlets growth. Methods Orthogonal array experimental design was applied to study the effects of TDZ, NAA, inositol on plantlets proliferation. Results Plantlets proliferation was significantly affected by TDZ and inositol. Optimal media to induce plantlets proliferation was Fast medium with TDZ 5.0 mg/L and NAA 0.05 mg/L and inositol 50 mg/L. and the proliferation multiple reached 7.3 times. Activated charcoal addition to culture medium promote growth of in vitro seedlings, and its suitable addition amount was 0.05 g/L. Conclusion The factors have significantly affected for seedlings proliferation in vitro ofBletilla striata. The most significant factor affecting seedlings proliferation in vitro of Bletilla striata was TDZ and inositol, and the general factor was NAA. The optimal medium can be applied to improve rapid propagation of Bletilla striata, which have the advantages of highly propagation coefficient, strong growth for plantlets in vitro.
