氯胺酮、咪唑安定与感染性休克大鼠肝HSP70的表达

摘    要:目的:观察氯胺酮,咪唑安定对感染性休克大鼠肝热休克蛋白70(HSP70)表达的影响。方法:健康成年SD大鼠88只,分为对照组(NS,E组)和试验组(EK,EKM组),NS组为正常对照组,E组给予内毒素20mg/kg,EK组,EKM组在同E组处理前分别给予氯胺酮80mg/kg,氯胺酮80mg/kg 咪唑安定0.5mg/kg。在A时点(内毒素处理后2h)和B时点(以内毒素组大鼠临终时为时点)测定肝HSP70表达及TNF-α血浆水平,并观察各组生存时间,结果:E组较NS组HSP70表达明显上调,TNF-α血浆水平明显升高(P<0.05),与E组比较,EK组HSP70表达明显上调,TNF-α血浆水平明显降低(P<0.05),大鼠存活率明显升高(P<0.05),EKM组与EK组比较各组数据无明显差异,结论:氨胺酮提高感染性休克大鼠肝HSP70的表达,抑制内毒素刺激的TNF-α的释放,提高氯胺酮的作用。α

Abstract:Objective:To observe the effect of ketamine,midazolam for the expression of heat shock proteins(HSP 70) in rat liver with septic shock. Methods:88 healthy and matured rats were distributed in random fashion into control groups (group NS,E)and test groups (group EK,EKM). Group NS served as normal control. Group E was treated with endotoxin 20mg/kg. Group EK,EKM were treated respectively with KTM80mg/kg. KTM80mg/kg + M 0.5mg/kg before receiving endotoxin 20mg/kg. The expression of HSP70 and TNF- α level were measured at 2 hours after endotoxin treatment and dying time for group E rats. Rats survival time were observed. Results:There was a significant (P<0.05) increase of HSP 70 and TNF- α level in the group E when compared with values of group NS. There was a significant(P<0.05) level in group EK when compared with values of group E. There was no difference concerning TNF-α level,HSP70 and survival rates between group EKM and group EK. Conclusions: Ketamine could increase the expression of HSP70 in rat liver with septic shock and survival rates and supree the release of TNF- α caused by endotoxin. Joint use of ketamine-midasolam did not affect the liver protection property of ketamine.
