苦参碱诱导人胃癌细胞凋亡及对端粒酶活性的影响

摘    要:目的 研究苦参碱对人胃癌细胞诱导凋亡作用及端粒酶活性变化。方法苦参碱处理人胃癌细胞株SGC-7901后,用四甲基偶氮唑蓝(MTT)法测定细胞生长抑制效应,显微镜下观察胃癌细胞形态变化,应用流式细胞仪(FCM)检测细胞周期和凋亡,用PCR-ELISA法对胃癌细胞端粒酶活性进行检测。结果不同浓度的苦参碱对SGC-7901有明显抑制作用,且呈时间依赖性及剂量依赖性,形态学观察也发现大量神经细胞的染色质凝聚,核碎裂,凋亡小体产生,细胞即出现明显的凋亡形态特征,FCM检测结果显示苦参碱作用后,G0/G1期细胞比例增高,S期、G2/M期细胞比例下降,苦参碱在诱导细胞凋亡的同时伴随端粒酶活性下调,且随苦参碱浓度增大和时间延长,抑制作用逐渐增强。结论苦参碱对人胃癌细胞具有明显抑制作用,诱导细胞凋亡并抑制端粒酶活性可能是其发挥抗癌作用的机制之一。

Abstract:Objective] To study inhibition effect of Matrine on the growth of gastric carcinoma cells SGC-7901. Methods] The growth inhibiting rate of SGC-7901 cell in vitro were measured by MTT assay. Apoptosis rate and cell cycle were measured by flow cytometry(FCM); Morphological changes were observed with fluorescence microscope. The activity of telomerase of SGC-7901 cell was examined by PCR-ELISA. Results] Different concentration of matrines were cytotoxic to SGC-7901 cell in time-dependent and dose-dependent manner. There were morphological changes including chromatin condensation, nuclear framentation and formed apoptotic bodies. The cell underwent apoptosis with typical morphological feature. The FCM analysis showed that the number of SGC-7901 cell of G0/G1 phase increased and the number of cell of S phase and G2/M phase decreased. At the same time, decreased telomerase activity was detected in matrine-induced apoptotic cell. Moreover, the inhibition effect on telomerase activity was enhanced in a dose-dependent and time-dependent pattern. Conclusion] Matrine is effective in growth inhibition on gastric carcinoma cell line. Matrine-induced apoptosis and inhibition on telomerase activity may be one of the anticancer machanisms.
