系统性红斑狼疮患者长链非编码RNA差异性表达研究

目的利用长链非编码RNA表达谱芯片技术筛选系统性红斑狼疮(SLE)患者的差异表达长链非编码RNA并对其进行分析,以进一步阐明系统性红斑狼疮发病的分子机制。方法分采用利用长链非编码RNA表达谱芯片检测技术,筛查系统性红斑狼疮及健康对照组单个核细胞中的差异表达长链非编码RNA,并通过Rreal-time PCR对部分差异表达基因进行验证。结果 uc003ngn、uc010loq、uc003wbg、uc003wax、HM-lincRNA1145、uc010jsn、uc003wax、HMlincRNA1145等lncRNA在SLE患者外周血单个核细胞中显著上调,AK022005、uc010cik、uc010gdp、uc010nwn、uc010imt、HMlincRNA678等lncRNA在SLE患者外周血单个核细胞中显著下调。结论多个长链非编码RNA在SLE患者外周血单个核细胞中异常表达,提示它们可能在SLE的发生发展中起着重要的调控作用。

Microarray-based long non-coding RNA expression profiles related to SLE

【Objective】 To screen differently expressed long non-coding RNA(lncRNA) in peripheral blood mononuclear cells between Systemic lupus erythematosus patients and health controls.【Methods】 The peripheral blood mononuclear cells of Systemic lupus erythematosus patients and health controls were screened for differently expressed long non-coding RNA by microarray.Then the result of microarray was verified by real-time PCR.【Results】 The upregulated lncRNA in SLE patients included uc003ngn,uc010loq,uc003wbg,uc003wax,Hmlinc RNA1145,uc010jsn,uc003wax and Hmlinc RNA1145,while the down-regulated lncRNA were AK022005,uc010cik,uc010gdp,uc010nwn,uc010imt and Hmlinc RNA678.【Conclusions】 The expression profile of long non-coding RNA is changed in SLE’s PBMC,which implies that those changed long non-coding RNA may be potential diagnosis biomarkers and play an important role in the pathogenesis of Systemic lupus erythematosus.
