实验性脑缺血再灌后迟发性神经元死亡表现为细胞凋亡

摘    要:目的:探讨脑缺血再灌后迟发性神经元死亡与细胞凋亡的关系。方法:采用脑缺血再灌损伤模型,观察缺血再灌后不同时期神经细胞损伤情况,免疫组化法分别检测缺血再灌后不同时期B的Bcl-2和Bax蛋白的表达,结果:沙鼠脑缺血再灌3d后,缺血神经细胞出现细胞凋亡改变(P<0.001;缺血再灌后6h及1d出现Bcl-2蛋白表达增加(P<0.01),而Bax蛋白则在再灌后6h至第7天呈现持续强阳性表达(P<0.001);结论:沙鼠短暂脑缺血再灌后,迟发性神经元残废表现为细胞凋亡,而Bax蛋白的高比率表达可能是导致细胞凋亡的重要因素之一。

Abstract:Objective:To explore the relation of delayed neuronal death with neuronal apoptosis following transient forebrain ischemia in the gerbil.Methods:Immunohistochemisty (IHC) were used to determine the expressions of the Bcl-2 and Bax proteins of the forebrain during different periods after post-ischemic reperfusion in the gerbil,and the neuronal damage was examined with microscope.Results:The neuronal apoptosis could be seen at the 3rd day of reperfusion,and most of the neurons showed apoptosis the 7th day of reperfusion (P<0.001) following 6min of ischemia;the Bcl-2 protein was only increased at the 6th hour and the 1st day of reperfusion (P<0.01),but the Bax protein was significantly increased durging the period from the 6th hour to the 7th day of reperfusion (P<0.001).Conclusions:The delayde neuronal death was apoptosis following transient ischemia in the gerbil;The persistant high expression of Bax protein may be an important factor that induces neuronal apoptosis after ischemia.
