SCID小鼠和BALB/c裸鼠对两株人体肿瘤细胞系移植敏感性的研究

摘    要:本文用MDA -MB2 31乳腺癌细胞致瘤块和人肺腺癌 1Ax - 83细胞系 ,在严重联合免疫缺陷 (SCID)小鼠、BALB c裸鼠的乳房垫和腋背侧皮下移植。结果表明 ,SCID小鼠对两株人体肿瘤细胞系成瘤率为 10 0 % (2 1 2 1)高于BALB c裸鼠 57 1% (8 14) ,两者差异显著 (P <0 0 5) ,SCID小鼠比BALB c裸鼠潜伏期明显缩短、且肿瘤生长较快。MDA -MB - 2 31乳腺癌细胞致瘤块移植组 ,SCID小鼠乳房垫接种者肺转移 10 0 % (5 5)、淋巴结转移 2 0 % (1 5) ,腋背侧皮下移植肺转移 50 % (2 4 )、淋巴结未见转移 (0 4 ) ;而BALB c裸鼠乳房垫接种者未见转移 (0 4 ) ,腋背侧皮下接种者仅 2 0 %(1 5)有肺转移。人肺腺癌 1Ax - 83细胞系移植组SCID鼠无自然消退 ,而BALB c裸鼠消退率 50 %(1 2 )。结论 :SCID小鼠比BALB c裸鼠更适宜做某些肿瘤移植模型 ,尤其是乳腺癌原位移植动物模型。

Abstract:Two human carcinoma cell lines, MDA-MB-231 breast cancer a nd LAX-83 lung adenocarcinoma cell lines were xenografted in scid (severe combin ed immunodeficiency mice) and BALB/c nude mice. MDA-MB-231 cells were subcutaneo usly injected into different parts (including breastpad and costal region) adn L AX-83 only one part (costal region) of the animals. Results showed that the tota l survival rates of two cell lines were 100% (21/21) in scid, 57.1%(8/14) in BAL B/c-nu. There was asignificant difference between two strains (P0.05). In the test of MDA-MB0231 transplantation, the Rates of lung metastasis were 100%(5/5), lymphatic metastasis 20%(1/5) with the breastpad injection, lung metastasis 50% (2/4) with costal injection in scid, and only 20% (1/5) of the lung metastasis c ould be observed with the costal injection in BALB/c-nu. In LAX-83 test, all of the carcinoma in scid did not disapear naturally. But 50% (1/20 did from BALB/c-nu. Studies suggested that scid was still better su ited for being an animal model of tumor experiments than BALB/c-nu, especially f or orthotopic transplantation of the breast cancers.
