摘    要:目的通过调整精卵交汇时间,提高C57BL/6J小鼠超数排卵的受精率。方法实验一以常规超排方法作为对照组,计算受精率;实验二验证C57BL/6J雄鼠受精能力;实验三在合笼后三个时间点检栓,确定超排鼠交配时间,计算精卵相遇时间;实验四将C57BL/6J雌鼠超排处理,停留6.5 h后配雄鼠,计算受精率,比较方法改进的效果。结果对照组取受精卵10.88枚/只;实验二取受精卵20.55枚/只,表明雄鼠的精子可以使卵子高比例受精,精子、卵子交汇时间是影响受精率的关键因素;实验三,在合笼后4 h雌鼠见栓鼠最多,确定对照组精卵交汇时间需7 h;根据以上实验结果,实验四将合笼时间推迟6.5 h,取受精卵22.27枚/只,比对照组提高了105%。结论调整精子、卵子交汇时间可显著提高C57BL/6J小鼠超数排卵受精率。

Abstract:Objective To explore the improving fertilization rate of superovulated C57BL/6J mice by adjusting sperm and egg encounter time. Methods Two hundred and forty-four 3-week-old female C57BL/6J mice and fifty 8-week-old male C57BL/6J mice were used in this study. (Ⅰ) To deterimine the fertilization rate by routine superovulation method, and use it as a control group. (Ⅱ) To verify the insemination ability of C57BL/6J male mice. (Ⅲ) To determine the time of coitus by checking vaginal plug at 3 time points after putting the female and male into one cage, and to calculate the time for sperm and egg encounter. (Ⅳ) Female C57BL/6J mice after superovulatory treatment, stayed for 6.5 h before mating with male mice. To collect fertilized eggs, to calculate the fertilization rate and compare the effects of different treatment methods. Results (Ⅰ) 10. 88 fertilized eggs per mouse were collected in the control group; (Ⅱ) In average 20. 55 fertilized eggs per mouse were collected in the second group. It was 88.87% higher than that of the control group, indicating the ability of male sperm to get a higher fertilization rate of ova, and the sperm and egg encounter time was the key factor of fertilization rate. (Ⅲ) For the majority of female mice showed vaginal plugs at 4 h after cohabitation, the sperm and egg encounter time in the control group could be estimated as approximately 7 h. (IV) Based on the above results, delayed the mating time for 6 h, the mean number of fertilized eggs per mouse was increased to 22. 27, by 105% increased than that of the control group. Conclusions Adjusting the sperm and egg encounter time can significantly increase the fertilization rate of superovulated C57BL/6J mice.
