食管癌p53基因突变的研究

应用非同位素ＰＣＲ－ＳＳＣＰ方法分析了３０例食管癌患者癌组织及相应癌旁组织Ｐ５３基因第５－９外显子，结果发现８例癌组织存在Ｐ５３基因突变。其中的４例为Ｐ５３基因杂合性丢失。部分癌旁组织检测到Ｐ５３基因突变，实验结果表明，Ｐ５３基因突变可能是分管癌癌变的早期遗传学损害，对食管癌的发生起重要作用。

Mutation of the p53 Gene in Esophageal Squamous Cell Carcinomas

30 cases of human squamous ccll carcinoma were examined by non -isotope PCR-SSCP for the presence of p53 mutations in exons 5, 6, 7, 8 and 9 in tumorous tissues and nontumorous tissues rcspcctlvely. Mutations werc detected at exons 5~8 of the p53 gene in 8 tumorous tissues as wcll as in some nontumorous tissues. Of all the eight cases that har boured p53 genc mutations, four wcre ohscrved to retaln LOH on the p53 locus. These results suggested that the mutation of the p53 gene could be an early genetic aberration in the car clnogenesis of squamous cell carc1nornas of the esophagus, and might play an important role in the occurrence of the tumor.
