摘    要:目的 了解不同浓度藏红花酸对HERG钾通道蛋白的影响.方法 采用Western blotting法、免疫荧光化学染色法和激光共聚焦显微镜检测法,检测不同浓度藏红花酸对稳定转染有HERG通道的HEK-293细胞中HERG钾通道蛋白表达的影响.结果 1、3、10和30 μmol·L-1藏红花酸对HERG钾通道蛋白的表达均无明显影响.结论 藏红花酸对HERG钾通道蛋白的表达无明显影响,作为抗心律失常药物成分具有较好的安全性.

Abstract:Objective To study the effect of different concentrations crocetin on HERG potassium channels protein expression.Methods Western Blotting,Immunofluorescence chemical staining method and laser confocal microsopy were used to detect the expression of HERG channel in the HERG-HEK cells with the influence of different concentrations of crocetin.Results Different concentrations of crocetin(1.0,3.0,10.0 and 30.0μM)were not significantly affected the expression of HERG potassium channels protein.Conclusions Crocetin had not significantly impact on the expression of HERG potassium channels protein,which provided a theoretical support for the safety of crocetin involving anti-arrhythmic mechanism as the herb as a Traditional Chinese Medicine used in clinical practice.
