酸枣仁汤对抑郁模型大鼠行为学和脑组织单胺类神经递质的影响

摘    要:目的:建立抑郁动物模型,观察酸枣仁汤对抑郁模型大鼠行为学和脑组织单胺类神经递质的影响,探讨其抗抑郁作用机理.方法:采用孤养结和慢性不可预见性温和刺激制备抑郁大鼠模型.通过体重增长变化、糖水消耗量和敞箱实验.评定酸枣仁汤对慢性应激抑郁模型大鼠的行为学改变,并用高效液相色谱法测定大鼠脑内单胺类神经递质的含量,运用统计学方法比较组间差异,说明酸枣仁汤对抑郁实验动物模型的影响.结果:模型大鼠出现了明显的行为学异常,脑内单胺类神经递质含量与正常组有显著差异,酸枣仁汤高剂量组可显著提高抑郁大鼠的糖水消耗量、敞箱实验评定、脑内单胺递质含量,其疗效与西药氯丙咪嗪相当.结论:孤养结合慢性不可预见性的温和刺激可制备比较理想的抑郁症模型,酸枣仁汤可显著改善抑郁模型大鼠的行为学异常,增加脑内单胺递质含量,且呈一定的量效关系.酸枣仁汤的抗抑郁作用可能与增加脑内单胺类神经递质含量有关.

Abstract:Objective:Set up the depression-model to observe the effect of Suanzaoren decoction on the depressive behavior and the monoamine neurotransmitters of DEP rat model,and throught it to probe the anti-depressive mechanism of Suanzaoren decoction.Methods:Isolated-living condition and chronic mild unpredictable stress are applied successively to set up DEP rat model.DEP rat is assessed by varitation of weight,Sucrose solution consumption test and Open-field test.They are used to estimate the change of behaviors of DEP rat model.High pressure liquid chromatography(HPLC)is applied to detect the monoamine neurotransmitters of the brains.Evaluating the efficiency of Suanzaoren decoction on the depressive behavioral tests and cerebral monoamine neurotransmitters of DEP rat model through statistics means.Results:The DEP rat model manifests significant changes on behaviors,the remarkable droping of cerebral monoamine neurotransmitters compared with the normal control group.High-dose group of Suanzaoren decoction augment the consumption of sucrose solution,besides improves the Open-field behaviors of DEP rat significantly.High-dose group of Suanzaoren decoction increase the cerebral monoamine neurotransmitters concentration of DEP rat significantly.Conclusions:Isolated-living condition and chronic mild unpredictable stress are applied successively to set up DEP rat model.Suanzaoren decoction can promote the depressive behaviors and monoamine neurotransmitters concentration of DEP rat model.The result present a dose-dependent effect.The anti-depressive mechanism of Suanzaoren decoction mainly is due to increasing the cerebral monoamine neurotransmitters concentration in the brains.
