三七总皂苷抑制HSC-T6增殖的实验研究

摘    要:目的研究三七总皂苷对HSC-T6细胞增殖的影响。方法将复苏后处于对数生长期的HSC-T6单细胞悬液稀释成浓度为2.0×10^5个/ml,并以0.1ml/孔接种,置于37℃、5%CO2培养箱内培养,24h后将不同稀释度的三七总皂苷按0.1ml/孔加入培养孔。实验另设培养液空白对照组和细胞对照组。每24h观察细胞形态及其生长状况,实验第4天取细胞上清液,所余细胞以MTT法用酶标仪测定OD值,计算出药物对细胞增殖的抑制率。结果三七总皂苷1/1、1/2、1/4、1/8、1/16、1/32、1/64、1/128倍比稀释浓度时的抑制率分别为83.9%、82%、83%、80.4%、85.3%、79.8%、70.7%、58.9%。1/128以下浓度组的抑制率逐渐降低。结论三七总皂苷对HSC-T6细胞的增殖有明显抑制作用,且有明显的浓度依赖,在128倍稀释浓度时仍有较高抑制率且细胞破坏较轻。

Study of hepatic-stellate-cell-T6 cell proliferation affected by total panax notoginseng sapoinins

Abstract:Objective to study the effect of Panax notoginseng sapoinins on proliferation of Hepatic-stellate-cell-T6 cell line.Methods After recovery,HSC-T6,in logarithmic growth,was diluted to a concentration of 2.0×10^5 /ml,and inoculated 0.1ml/hole in incubator at 37℃,saturated humidity and 5% CO2.24h later,Panax notoginseng sapoinins in different concentrations were added by 0.1ml /hole,Comparing with tank control group and cell control group.Cell morphology and their growth condition were observed every 24h.On 4th day,cell supernatant were collected,and OD value of the remaining cells was measured by MTT,and the inhibitory ratio of cell proliferation was calculated.Results The inhibitory rate of Panax notoginseng sapoinins in concentrations of 1/1,1/2,1/4,1/8,1/16,1/32,1/64,1/128 were 83.9%,82%,83%,80.4%,85.3%,79.8%,70.7%,58.9% respectively,and the inhibitory rate decreased gradually for the groups in concentrations of lower than 1/128 times.Conclusion Panax notoginseng sapoinins markedly inhibited the cell proliferation of HSC-T6,with an obvious concentration-dependent,and their dilution of 128 times still had a higher inhibitory rate and destroyed cells less.
