肝癌组织中nm23-H1、PTEN的表达及其临床病理的意义

摘    要:目的探讨nm23-H1、PTEN在肝细胞癌中的表达及其与患者的临床病理特点的关系。方法应用免疫组化S-P方法,检测50例肝癌组织中nm23-H1、PTEN的表达,分析其与临床病理之间的关系。结果 nm23-H1、PTEN蛋白阳性表达为棕黄色,定位于细胞浆。10例正常肝组织和50例癌旁肝组织均可见阳性表达。肝癌组织nm23-H1、PTEN阳性率表达分别为60%(30/50)、54%(27/50),明显低于癌旁肝细胞阳性率表达100%(50/50,P〈0.001,P〈0.001)。nm23-H1和PTEN表达与肝癌病理分级、临床分期、肝内转移密切相关,而与患者性别、年龄、肝癌癌灶大小、肝包膜有无、甲胎蛋白水平无明显相关性。结论肝癌组织中nm23-H1、PTEN表达减低与肝癌发生发展有相关性。检测nm23-H1、PTEN两种蛋白在肝癌组织中的表达能反映肝癌的恶性程度及侵袭转移能力。

Expressions of nm23-H1 and PTEN in hepatocellular carcinoma tissues and their clinicopathological significance

Abstract:Objective To explore the expressions of nm23-H1 and PTEN in primary hepatocellular carcinoma(HCC)and their clinicopathological significance.Methods Expressions of nm23-H1 and PTEN protein in liver cancerous tissues and paired para-cancerous tissues of 50 HCC patients were detected by immunohistochemistry.The relationship between the expression levels of nm23-H1、PTEN protein and the clinicopathological parameters of the HCC patients was analyced.Results nm23-H1 and PTEN protein were positively stained in cytoplasm in all of the 10 normal liver tissues and the para-carcinomal tissues of the 50 HCC patients,while the positive rates of nm-23-H1 and PTEN in liver cancerous tissues were 60%(30/50 cases)and 54%(27/50 cases)respectively,which were significantly lower than those of the para-carcinomal tissues(P〈 0.001,P〈 0.001,respectively).Furthermore,the down-regulations of nm23-H1 and PTEN proteins in HCC tissues were doesly related to the histological Edmondson grading,clinical stages,and intrahepatic metastasis,while no relation between their expressions and the gender,age,tumor size,liver capsule existing or AFP expression level was observed.Conclusions The reduced expressions of nm23-H1 and PTEN might contribute to tumor development and metastasis in HCC.Examination of protein expressions of nm23-H1 and PTEN in HCC may help to understand the malignance degree and the potential metastasis of the tumor.
