造影剂和甘露醇对大鼠肾小球内皮细胞影响的比较研究

摘    要:目的研究非离子型低渗造影剂和等渗甘露醇对体外培养大鼠肾小球内皮细胞的损伤情况,分析造影剂毒性作用与其高渗性的关系。方法用MTT法检测同等渗透压的碘佛醇(32mgI/ml,100μl/m1)和甘露醇对大鼠肾小球内皮细胞增殖的影响,并用化学发光法检测肾小球内皮细胞乳酸脱氢酶(LDH)的释放;Annexin V/PI染色分析二者诱导肾小球内皮细胞凋亡的情况。结果非离子型低渗造影剂碘佛醇和甘露醇都能抑制大鼠肾小球内皮细胞增殖,但碘佛醇的作用更显著(P〈0.01);碘佛醇作用24小时后引起的肾小球内皮细胞LDH释放增加具有统计学意义(P〈0.01),甘露醇组与对照组并没有显著性差异;而碘佛醇诱导细胞凋亡的作用更显著。结论非离子型低渗造影剂与同等渗透压的甘露醇相比,损伤肾小球内皮细胞和诱导凋亡作用更为明显,造影剂的细胞毒性可能还与渗透压之外的其它因素有关。

Effects of contrast media and mannitol on rat glomerular endothelial cells

Abstract:Objective To investigate the effects of non-ionic low osmolar contrast medium(LOCM)and equal osmolar mannitol on injury of rat glomerular endothelial cells. To analyze the relationship between LOCM and its hyperosmolality. Methods The effect of LOCM on rat glomerular endothelial cells was compared with equal osmolar mannitol with MTT assay. The release of lactate dehydrogenase release (LDH) from glomerular endothelial cells was detected with chemiluminescence. Apoptosis was determined with annexin V/PI stain. Results The cell viability decreased in in the glomerular endothelial ceils treated with ioversol and mannitol, but the former was more serious than the later one. (P 〈0. 01). The release of LDH in glomerular endothelial cells after 24 hours exposure to ioversol increased significantly. The apoptosis induced hy ioversol was more obvious than that by mannitol. Conclusions Compared with equal osmolar mannitol, LOCM can obviously induee injury and apoptosis of rat glomerular endothelial cells.
