摘    要:目的:检测病理性瘢痕组织中凋亡抑制蛋白Livin和促凋亡因子Smac的表达情况及两者之间的关系。方法:分别采用RT-PCR和免疫组织化学方法检测55例病理性瘢痕(25例瘢痕疙瘩、30例增生性瘢痕),24例非病理性瘢痕和20例正常皮肤组织中Livin和SmacmRNA和蛋白的表达。结果:以上4种组织中Livin和SmacmRNA和蛋白的表达差异均有统计学意义(mRNA:F=269.533和95.514,P<0.001;蛋白:χ2=36.611和26.320,P<0.001);瘢痕疙瘩、增生性瘢痕中LivinmRNA的表达水平和蛋白的阳性表达率高于非病理性瘢痕及正常皮肤组织,SmacmRNA的表达水平和蛋白的阳性表达率低于非病理性瘢痕及正常皮肤组织(P<0.05或0.008)。病理性瘢痕组织中Livin和Smac蛋白的表达呈负关联(rP=0.418,P<0.001)。结论:Livin和Smac可能在病理性瘢痕凋亡信号传导网络中起重要的作用。

Abstract:Aim:To investigate the expressions of Livin and Smac in pathologic scar tissue.Methods:RT-PCR and SP immunohistochemical methods were used to detect the mRNA and protein expressions of Livin and Smac in 55 pathologic scar(25 cases of keloid,30 cases of hypertrophic scar),24 cases of non-pathologic scar and 20 cases of normal skin tissue.Results:There were significant differences in the expressions of Livin and Smac mRNA and protein in four kinds of tissue(mRNA:F=269.533 and 95.514,P<0.001;protein:χ2=36.611 and 26.320,P<0.001).Compared with the non-pathologic scar group and normal skin tissue group,the mRNA expression level and protein positive expression rate of Livin in keloid group and hypertrophic scar group were higher,while the mRNA expression level and protein positive expression rate of Smac in keloid group and hypertrophic scar group were lower(P<0.05 or 0.008).There was negative association between Livin and Smac protein expression in pathologic scar(rP=0.418,P<0.001).Conclusion:Livin and Smac might play an important role in the apoptosis pathway of pathologic scar.
