食管癌组织中KAI1基因的表达

摘    要:目的探讨KAI1基因的表达与食管癌浸润转移的关系.方法采用RT-PCR及免疫组化方法,检测35例食管癌组织及其正常黏膜中KAI1 mRNA及蛋白的表达.结果在食管癌组织及其正常黏膜中,KAI1 mRNA的表达差异无统计学意义(P>0.05),而癌组织中KAI1蛋白的表达明显低于其正常黏膜(P<0.05),但KAI mRNA及蛋白表达均与肿瘤的分化程度、浸润深度及淋巴结转移无关(P>0.05).结论食管癌癌变可能与KAI1蛋白的低表达有关,但与KAI1 mRNA的表达无关,推测这可能是由于翻译后修饰所致.

Abstract:Aim:To investigate the relation between the expression of KAI1 gene and the invasion and metastasis of esophageal carcinoma. Methods:The expression of KAI1 mRNA was detected by RT PCR and the KAI1 prottin was detected by immunohistochemical method (SABC) in 35 cases of esophageal cancer tissues and its normal mucosa tissues. Results: The expression of KAI1 mRNA showed no difference between esophageal cancer tissues and normal mucosa,but the expression of KAI1 protein were significantly lower in cancer tissues than that in normal mucosa tissues( P <0.05). Neither the expression of KAI1 mRNA or that of KAI1 protein appeared to be correlated with lymph node metastasis,the depth of invasion and differentiation of cancer tissues. Conclusion: Esophagus carcinogenesis may be related to the reduced expression of the KAI1 protein but not to the level of mRNA,which may result from posttranslational modifications.
