摘    要:目的:检测食管癌和癌旁正常组织Ras相关区域家族1A(RASSF1A)基因启动子区甲基化变化.方法:采用甲基化特异性聚合酶链反应(MSP)检测79例食管鳞癌患者癌组织及对应的20例癌旁正常组织RASSF1A基因启动子区甲基化,并观察食管癌组织RASSF1A基因启动子区甲基化与临床病理变化的关系.结果:79例食管鳞癌组织RASSF1A基因启动子区甲基化阳性率(67%,53/79)明显高于癌旁正常组织(15%,3/20).食管癌组织中不同年龄、分化程度的RASSF1A基因启动子区甲基化阳性率差异有统计学意义(P<0.05),而淋巴结转移与否、肿瘤分期间甲基化阳性率差异无统计学意义.20例同一个体癌组织甲基化阳性率明显高于癌旁正常组织(50%(10/20)vs 15%(3/20))(P<0.05),仅3例同时出现甲基化,一致率为15%.结论:RASSF1A基因启动子区甲基化是食管鳞癌频发的分子事件.食管鳞癌组织RASSF1A基因甲基化明显高于癌旁正常组织,并且和年龄、分化程度有关.

Abstract:Aim: To detect the promoter methylation of RASSF1A gene in esophageal cancer and adjacent normal tissue.Methods:Methylation special PCR was applied to detect RASSF1A methylation on 79 cases of esophageal squamous cell carcinoma (SCC) and 20 cases of adjacent normal tissue.Results: The detection rate of RASSF1A methylation in SCC (67%, 53/79) was much higher than that in adjacent normal tissue (15%, 3/20). The methylation of RASSF1A gene had significant difference among different age groups and differentiation degree, P<0.05. However, the RASSF1A methylation was not associated with lymph node metastasis or tumor staging. The methylation rate in SCC was apparently higher than that in adjacent normal tissue from the same patient (50%(10/20) vs 15%(3/20)), P<0.05. Conclusion: RASSF1A methylation is a common event in SCC; methylation in SCC is much higher than that in adjacent normal tissue and correlated with the age and differentiation of the patients.
